摘  要:目的采用HPLC法同时测定温郁金须根挥发油中莪术二酮、莪术醇、吉马酮、呋喃二烯等4种成分的含量,比较温州瑞安5个产区温郁金须根中4种挥发油成分的含量差异。方法采用Agilent Zorbax SB-C_(18)柱,以乙腈-水为流动相,梯度洗脱,流速1 mL·min^(-1),柱温30℃,检测波长216 nm,进样量5μL。结果莪术二酮、莪术醇、吉马酮、呋喃二烯的线性范围分别为8.88~222.0、8.72~218.0、17.92~448.0、9.20~115.0μg·mL^(-1),平均回收率分别为99.19%、101.16%、103.76%、96.44%,RSD均小于3.0%。温郁金须根中挥发油的含量以霞岙村最低(0.12%),五仁山村最高(0.95%),挥发油中莪术二酮、莪术醇的含量以五仁山村最高,分别为2.74%、1.34%;吉马酮、呋喃二烯的含量以沙洲村最高,分别为0.88%、4.92%。结论所用方法快速、准确,适用于温郁金须根中挥发油成分的含量测定。不同产区温郁金须根的挥发油得率及主要成分含量差异较大,需建立温郁金的生产质量管理规范,保证其品质和道地性。OBJECTIVE To determine the contents of curcumin,curcumol,germacrone and furadiene in the essential oil from the fibrous root of Curcuma wenyujin by HPLC,and to compare the differences of the main components in the essential oil from the fibrous roots of C.wenyujin from five producing areas in Ruian,Wenzhou.METHODS The column was Agilent Zorbax SB-C_(18)and acetonitrile water was used as mobile phase for gradient elution with the volume flow rate of 1 mL·min^(-1),the column temperature of 30℃,the detection wavelength of 216 nm,and the injection volume of 5μL.RESULTS The linear ranges of curdione,curcumol,gemacrone and furadiene were 8.88-222.00μg·mL^(-1),8.72-218.00μg·mL^(-1),17.92-448.00μg·mL^(-1)and 9.20^(-1)15.00μg·mL^(-1),respectively.Their average recoveries were 99.19%,101.16%,103.76%and 96.44%with RSDs less than 3.0%.The lowest content of essential oil in the roots of C.wenyujin in Xia’ao village was 0.12%,while the highest in Wurenshan village was 0.95%,where the highest contents of curdione and curcumol in the essential oil were 2.74%and 1.34%,respectively.The highest content of gemmazone and furandiene respectively was 0.88%and 4.92%in Shazhou village.CONCLUSION The method is rapid and accurate,which is suitable for the determination of four main components in the essential oil of C.wenyujin.The contents of essential oil and main components in the fibrous root of C.wenyujin in different producing areas were quite different,so it is necessary to establish the production quality management standard of C.wenyujin to ensure its quality and authenticity.
