摘 要:目的以正交设计试验优选元胡止痛方的最佳水提取工艺,为元胡止痛方的提取工艺条件以及工业化生产提供实验依据。方法将影响提取效果的A(浸泡时间)、B(提取溶剂量)、C(提取时间)以及D(提取次数)4个主要因素,每个设3个水平,以正交表L_(9)(3^(4))设计正交实验;通过梯度洗脱并以HPLC法同时测定指标成分延胡索乙素、欧前胡素的含量,经加权评判进行提取工艺的优选。结果加权后元胡止痛方提取工艺影响因素由大到小顺序依次是B>C>A>D;B和C起主要作用,其中B有极显著性差异(P<0.01);C有显著性差异(P<0.05),而A、D均无统计学意义。结论考虑到经济效益,确定元胡止痛方以10倍量水浸泡30 min后再回流提取2次,每次提取3 h为其最佳提取工艺。经验证表明,该工艺稳定,可行,可考虑作为工业化生产条件。OBJECTIVE To optimize the optimal water extraction process of Yuanhu Zhitong Recipe by orthogonal design experiment,and to provide experimental basis for the extraction process conditions and industrial production of Yuanhu Zhitong Recipe.METHODS Set the four main factors A(soak time),B(extraction solvent amount),C(extraction time)and D(extraction times)that affect the extraction effect to 3 levels each.Orthogonal table L_(9)(3^(4))was used to design orthogonal experiments;by gradient elution and simultaneous determination of the content of the index components dl-tetrahydropalmatine and imperatorin by HPLC method,the extraction process was optimized by weighted evaluation.RESULTS The influencing factors of the extraction process of Yuanhu Zhitong Recipe after weighting were B>C>A>D in order;B and C played a major role,in which B had a very significant difference(P<0.01);C had a significant difference(P<0.05),while A and D had no statistical significance.CONCLUSION Considering the economic benefits,it is determined that Yuanhu Zhitong Recipe is immersed in 10 times the amount of water for 30 minutes and then refluxed for extraction twice,each extraction for 3 hours is the best extraction process.The verification shows that the process is stable and feasible,and can be considered as industrial production conditions.
