摘 要:目的探讨菊苣酸对心肌缺血再灌注(ischemia reperfusion,I/R)小鼠心肌损伤、氧化应激及炎性反应的影响.方法构建心肌I/R的BALB/c小鼠模型.40只小鼠随机分为5组:对照组,I/R模型组,菊苣酸低、中、高剂量组.HE染色观察小鼠心肌组织损伤情况.ELISA检测心肌肌钙蛋白Ⅰ(cTnⅠ)、肌酸激酶同工酶(CK-MB)、肌红蛋白(Mb)、TNF-α,IL-6和IL-10水平.TUNEL检测心肌组织细胞凋亡.试剂盒检测SOD、MDA及LDH水平.蛋白印迹检测Bax、Bcl-2及c-Myc表达.结果与对照组比较,I/R模型组心肌组织损伤严重,cTnⅠ、CK-MB、Mb、MDA、LDH、TNF-α、IL-6水平和细胞凋亡率升高(P<0.05),Bax/Bcl-2表达上调(P<0.05),c-Myc表达下调(P<0.05),SOD、IL-10水平降低(P<0.05).与I/R模型组比较,菊苣酸中、高剂量组心肌组织损伤减轻,cTnⅠ、CK-MB、Mb、MDA、LDH、TNF-α、IL-6水平和细胞凋亡率降低(P<0.05),Bax/Bcl-2表达下调(P<0.05),c-Myc表达上调(P<0.05),SOD、IL-10水平升高(P<0.05).结论菊苣酸通过调节心损指标、氧化应激标记物及炎性因子,从而减轻心肌I/R小鼠的心肌损伤,对心脏起到保护作用.Objective To investigate the effects of chicoric acid on myocardial injury,oxidative stress and inflammatory response in myocardial ischemia reperfusion(I/R)mice.Methods BALB/c mouse model of myocardial I/R was established.Forty mice were randomly divided into 5 groups:control group,I/R model group,low-,medium-and high-dose chicoric acid groups.HE staining was used to observe the damage of myocardial tissue in mice.The levels of myocardial troponin I(cTn I),creatine kinase isoenzyme(CK-MB),myoglobin(MB),TNF-α,IL-6 and IL-10 were detected by ELISA.TUNEL was used to detect apoptosis of myocardial cells.The levels of SOD,MDA and LDH were detected.Western blotting was used to detect the expression of Bax,Bcl-2 and c-Myc.Results Compared with the control group,the I/R model group showed myocardial tissue damage,increased levels of cTn I,CK-MB,MB,MDA,LDH,TNF-α,IL-6 and increased apoptosis rate(P<0.05),up-regulated expression of Bax/Bcl-2(P<0.05),and down-regulated expression of c-Myc(P<0.05),and reduced SOD and IL-10 levels(P<0.05).Medium-and high-dose chicoric acid could ameliorate the myocardial tissue injury,decrease the levels of cTn I,CK-MB,MB,MDA,LDH,TNF-α,IL-6 and apoptosis rate(P<0.05),down-regulate expression of Bax/Bcl-2(P<0.05),up-regulate the expression of c-Myc(P<0.05),and increase the levels of SOD and IL-10(P<0.05).Conclusion Chicoric acid can reduce myocardial injury in I/R mice and protect the heart by regulating cardiac damage indexes,oxidative stress markers and inflammatory factors.
