摘 要:抗菌药物的不合理使用,造成了细菌耐药性暴发和药物残留增加,这不仅降低了畜牧业生产,而且危害着人类的身体健康。制定合理健全的药物剂量给药方案能够缓解抗菌药物使用所面临的巨大压力,而药动学和药效学(PK-PD)模型能够对作用时间、药物浓度和治疗效果之间的内在关系进行拟合和定量分析,更加客观和科学地评价药物在体内的动态变化过程和作用效果。它在抗菌药物的合理使用,减少药物的耐药性产生和新药物的研发方面发挥重要作用。基于前人的研究,对PK-PD模型在兽用抗菌药物给药方案制定及优化方面的应用进行综述,为抗菌药物的使用提供参考依据。The unreasonable use of antimicrobials generally leads to the outbreak of bacterial drug resistance and the increase of drug residues,which not only reduces the production of animal husbandry,but also endangers the health of human beings.A reasonable and sound drug dose administration scheme must be established,which can alleviate the huge pressure faced by the use of antibacterial drugs.And the internal relationship among time,drug concentration and therapeutic effect is fitted and quantitatively analyzed by pharmacokinetic-pharmacodynamic(PK-PD)model,and the dynamic process and effect of drugs in vivo are evaluated more objectively and scientifically,which plays an important role in the rational use of antimicrobials,reducing drug resistance and the research and development of new drugs.Based on previous studies,the application of the PK-PD model in the formulation and optimization of veterinary antibacterial drug dosing schedules was discussed,which can help provide a reference for the research of veterinary antibacterial drugs.
