摘 要:采用锥形瓶液体培养方式,以广叶绣球菌(Sparassis latifolia)为试验材料,通过考察菌丝生物量及菌球大小评分,研究培养时间、摇瓶转速、培养温度、培养液起始pH及摇瓶装液量对绣球菌液体菌种制备质量的影响。单因素综合试验结果为,适宜培养条件为培养时间12天,转速210 r/min,培养温度26℃,pH 3.5,装液量75 mL,该条件下所形成的菌丝球均匀细小,菌丝生物量大,菌丝萌发点多,是良好的液体菌种材料。To meet the purpose of liquid spawn application in large-scale edible mushroom production,taking dry weight of mycelial biomass and mycelial ball diameter score as response value,the study was carried out to optimize the effects of liquid culture conditions(incubation time,temperature,rotational speed,initial pH value and volume of medium in flask)on the quality of Sparassis latifolia liquid spawn.The results showed that the optimum quality of liquid spawn could be attained along with evenly small mycelial ball,high mycelial biomass and abundant development under the incubation conditions of incubation time 12 d,rotational speed 210 r/min,temperature 26℃,initial pH value 3.5,and the volume of medium 75 mL.
