摘  要:本文以半枫荷(Semiliquidambar cathayensis)不同组织为研究对象,采用超高效液相色谱串联四级杆飞行时间质谱(UHPLC-QTOF/MS)代谢组学手段检测其化学成分种类,通过主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)等方法找出各组织间的差异代谢物,依据精确分子质量测定和相关文献信息对差异代谢物进行鉴定。结果显示:半枫荷叶片、茎和根中共检测到169个代谢物,结合PCA法综合判别出38个显著差异代谢物。差异代谢物在不同组织中表达模式不同:叶片中表达上调的差异代谢物较多,其次是根,再次是茎。不同组织中主要共有差异代谢物有5个,分别为柠檬酸、山奈酚3-O-桑布双糖苷、3-β-羟基齐墩果酸丁二酸单酯、1-O-乙酰基-α-麦芽糖、胞苷;叶片与根、茎相比主要差异代谢物有17个,主要为黄酮类、萜类和多糖类化合物;茎与叶片、根相比主要差异代谢物有4个,包括毛蕊花糖苷和β-熊果苷;根与叶片、茎相比主要差异代谢物有4个,包括香草酸、山茶苷B和三癸酸甘油酯。半枫荷不同组织中差异萜类化合物主要为三萜化合物,齐墩果酸等三萜化合物是区分半枫荷不同组织的重要差异性物质;不同种类的黄酮类化合物在半枫荷不同组织分布情况也不相同,大部分差异黄酮类化合物在叶片中富集明显。In this paper,an ultra-high performance liquid chromatography with quadrupole time-of-flight mass spectrometry(UHPLC-QTOF/MS)–based plant metabolomics approach was established to detect the chemical composition of different tissues of Semiliquidambar cathayensis.Through principal component analysis(PCA),orthogonal partial least squares discriminant analysis(OPLS-DA),and other methods,the metabolites between different tissues were identified based on accurate molecular mass determination and relevant literature information.The results show that a total of 169 metabolites were detected in the leaves,stems and roots of S.cathayensis,and 38 significantly different metabolites were comprehensively identified in combination with PCA.Metabolites had different expression patterns between leaves,stems and roots,and more metabolites were up-regulated in leaves,followed by roots,and then stems.There are five main metabolites in different tissues,namely citric acid,kaempferol 3-O-sambubi glycoside,3-β-hydroxyoleanolic acid succinate monoester,1-O-acetyl-α-maltose,and cytidine.Compared with roots and stems,leaves have 17 metabolites,mainly flavonoids,terpenoids,and polysaccharide compounds.Compared with leaves and roots,stems have four main metabolites,including verbascoside andβ-arbutin.Compared with leaves and stems,roots have four main metabolites,containing vanillic acid,camelliaside B and tridecanoic acid.The terpenoids in different tissues are mainly triterpenoids.These triterpenoids,such as oleanolic acid,are the most important distinguishing tissue features.There is a variety of flavonoids in different tissues,and most of the flavonoids are obviously enriched in leaves.
