摘 要:以GH35(glyco_hydro_35,PF01301)结构域保守序列为种子序列,在基因组范围鉴定了苹果(Malus domestica)等6种植物的β-半乳糖苷酶(β-galactosidases,BGals)基因家族,并对苹果BGals(MdBGals)进行了生物信息学和表达模式分析。结果表明,苹果基因组存在23个BGal基因,其氨基酸序列数、分子质量和等电点分别介于163~924、18.99~103.57 kDa和5.14~9.05之间,主要位于细胞壁。在不同植物中BGals相对保守,但在部分植物类群中存在较快的进化速率。苹果BGals启动子区域存在多个响应脱落酸、茉莉酸甲酯和水杨酸等激素信号和防卫信号的顺式作用元件。接种腐烂病菌后,苹果韧皮组织中多个BGals呈下调表达的趋势,也有少数成员上调表达。MD02G1079200、MD15G1306800和MD10G1301600表达量的变化较为明显,可作为进一步研究苹果抗腐烂病的候选基因。Using the conserved sequence of GH35(glyco_hydro_35,PF01301)domain as seed sequence,the gene family ofβ-galactosidase(BGals)was identified in six plants including apple(Malus domestica)and so on.And the bioinformatics and expression pattern of BGals in apple(MdBGals)were analyzed.The results showed that there were 23 BGal genes in apple genome.23 BGals were identified in the apple genome,with amino acid size(AA),the molecular weight(kDa),and isoelectric point varying from 163 to 924,18.99 to 103.57,and 5.14 to 9.05,respectively,which were mainly located in cell wall.BGals are relatively conservative in different plants,but there is a rapid evolution rate in some plant groups.There are many cis-acting rgulatory elements(cis-element)in the promoter region of apple BGals that respond to hormone and defense signals such as abscisic acid(ABA),methyl jasmonate(MeJA)and salicylic acid(SA).During Vm infection,most differently expressed BGals showed a down-regulation pattern,and a few of them were up-regulated.The expression levels of MD02G1079200,MD15G1306800 and MD10G1301600 changed significantly,which could be used as candidate genes for further study.
