摘 要:目的:通过原代培养神经元细胞,探究糖皮质激素受体(GR)功能对单胺氧化酶A(MAO A)的影响及其可能通路。方法:取生后48 h内的SD大鼠前额叶,经反复消化沉淀后,对神经元进行纯化并培养,48 h后,分为地塞米松(DEX)组、米非司酮(mifepristone,RU486)组和对照组,分别予以GR激动剂DEX、GR抑制剂RU486和等体积相同溶剂干预,继续培养48 h。采用免疫细胞化学染色法检测大鼠神经细胞Sp1、Krüppel样转录因子-11(KLF11)和MAO A表达情况。结果:DEX干预后,前额叶神经细胞中KLF11、Sp1及MAO A的表达均较对照组升高,而RU486干预后,Sp1和MAO A的表达较对照组降低,但KLF11的表达无明显变化。结论:糖皮质激素(GC)可提高前额叶区GR的表达;GC可通过激活GR,提高脑内神经细胞KLF11、Sp1和MAO A表达水平,Sp1的表达和MAO A存在正相关,提示GR功能与MAO A之间可能通过KLF11、Sp1起交互作用。Objective:To explore the effect of the glucocorticoid receptor(GR)on monoamine oxidase A(MAO A)expression and detect the biological pathways involved.Methods:Neurons in the prefrontal cortex of SD rats postnasal 48 hours were extracted and cultured for 48 hours and randomly separated into dexamethasone(DEX)mifepristone(RU486)group and control group.After EDX,RU486 and same valume medium were administrated,neurons were cultured for another 48 hours.Immunocytochemical technique and image analysis with integrated optical density(IOD)as an index were adopted to evaluate the expression level of KLF11,Sp1,and MAO A.Results:The expression level of KLF11,Sp1,and MAO A in prefrontal neural cells administered with GR agonist(DEX)was significantly higher than that in the control group,and the expression of Sp1 and MAO A was significantly decreased after intervention with GR antagonist(RU486),while no significant difference was found between the two groups in terms of the expression of KLF11.Conclusion:Glucocorticoid can increase the expression level of GR as well as KLF11,Sp1,and MAO A in prefrontal cortex neurons,and there is a positive correlation between the expression of Sp1 and MAO A,which indicates that the interaction between the GR and MAO A may be realized through KLF11 and Sp1.
