摘  要:生长停滞特异性基因6(GAS6)/TAM信号通路在血小板的充分、持续活化以及血栓稳定中起着至关重要的作用。抑制GAS6/TAM通路可在不增加内出血的前提下,减少血小板聚集、改变构象、收缩血凝块,以及抑制血小板在流动条件下的聚集和活化标志物在表面的表达等预防动静脉血栓形成。与野生型小鼠相比,GAS6基因缺陷小鼠和TAM受体基因缺陷小鼠在体内表现出对动脉和静脉血栓形成的保护,但不表现出自发性或长时间出血,且在使用GAS6-TAM信号通路抑制剂的野生型小鼠中也观察到了类似结果。由此推断GAS6/TAM信号通路可能在适度降低血小板活化和减少血栓形成的同时,仍保留了血小板聚集所起到的重要止血功能。未来对GAS6/TAM信号通路抑制剂的研究可能有助于改善血栓性疾病的治疗。The growth arrest-specific gene 6(GAS6)/TAM signaling pathway plays a vital role in the full and continuous activation of platelets and the stability of thrombus.Inhibition of the GAS6/TAM pathway can reduce platelet aggregation,change conformation,shrink blood clots,and platelet aggregation under flow conditions and surface expression of activation markers without increasing internal bleeding,thereby prevent arteriovenous thrombosis.Compared with wild-type mice,GAS6 gene-deficient mice and TAM receptor gene-deficient mice showed protection against arterial and venous thrombosis in vivo,but did not show spontaneous or prolonged bleeding,and similar results were observed in wild-type mice using GAS6-TAM pathway inhibitors.So,it can be inferred that the GAS6-TAM pathway may moderately reduce platelet activation and reduce thrombosis,while still retaining the important hemostatic function by platelet aggregation.Future research on GAS6/TAM pathway inhibitors may help to improve the treatment of thrombotic diseases.
