摘  要:目的研究黄芪及其发酵物对甲状腺功能减退症大鼠肾损伤和氧化应激的影响及其作用机制。方法将80只Wistar大鼠依据随机数字法分为正常对照组(10只)和造模组(70只),采用化学诱导法(0.05%丙硫氧嘧啶自由饮水)复制甲状腺功能减退症大鼠模型,造模成功后,造模组依据随机数字法分为甲状腺功能减退症模型组、黄芪低剂量组、黄芪中剂量组、黄芪高剂量组、黄芪发酵物低剂量组、黄芪发酵物中剂量组、黄芪发酵物高剂量组,每组10只。然后给予不同剂量的黄芪及其发酵物治疗4周后处死大鼠,测定各组大鼠血清超氧化物歧化酶活性和丙二醛水平,以及大鼠肾脏功能指标血清肌酐、尿素氮水平,肾组织切片苏木精-伊红染色观察肾脏的病理情况。结果与正常对照组比较,甲状腺功能减退症模型组大鼠超氧化物歧化酶活性明显降低[(243.06±6.38)kU/L比(269.30±4.02)kU/L](P<0.01),丙二醛水平明显增加[(6.82±0.55)μmol/L比(5.53±0.20)μmol/L](P<0.05),尿素氮和肌酐水平明显升高[(5.07±0.15)mmol/L比(3.62±0.23)mmol/L、(26.22±0.36)μmol/L比(14.73±0.24)μmol/L](P<0.01);与甲状腺功能减退症模型组比较,黄芪中、高剂量组和黄芪发酵物中、高剂量组超氧化物歧化酶活性显著升高(P<0.01),丙二醛、尿素氮和肌酐水平均明显降低(P<0.05或P<0.01),病理情况有所改善。结论黄芪及其发酵物可改善甲状腺功能减退症大鼠肾损伤,其作用机制与提高甲状腺功能减退症大鼠的抗氧化能力有关。Objective To study the effects of astragalus mongholicus and its fermentative matters on kidney injury and oxidative stress of rats with hypothyroidism,and its mechanism of action.Methods Eighty Wistar rats were divided into a normal control group(10)and a model-building group(70)according to random number method.Chemical induction method was adopted in this study(0.05%propylthiouracil,drinking water freely)to replicate hypothyroidism rat model.After building models successfully,the model group was further divided by the random number method:a hypothyroidism model group,an astragalus mongholicus low-dose group,an astragalus mongholicus medium-dose group,an astragalus mongholicus high-dose group,an astragalus mongholicus fermentative matters low-dose group,an astragalus mongholicus fermentative matters medium-dose group,and an astragalus mongholicus fermentative matters high-dose group,10 in each group.The rats were treated by different doses of astragalus mongholicus or its fermentative matters for 4 weeks and then killed.And then the activity of superoxide dismutase and malondialdehyde level in the serum,as well as the renal function indexes of levels of creatinine and urea nitrogen,in each group were measured,plus the pathological changes of kidney were observed by hematoxylin-eosin staining in renal tissue sections.Results Compared with the normal control group,the activity of superoxide dismutase in the hypothyroidism model group was significantly decreased[(243.06±6.38)kU/L vs(269.30±4.02)kU/L](P<0.01),the malondialdehyde level was significantly increased[(6.82±0.55)μmol/L vs(5.53±0.20)μmol/L](P<0.05),and the urea nitrogen and creatinine levels were significantly increased[(5.07±0.15)mmol/L vs(3.62±0.23)mmol/L,(26.22±0.36)μmol/L vs(14.73±0.24)μmol/L](P<0.01).Compared with the hypothyroidism model group,the activities of superoxide dismutase in the astragalus medium and high dose groups and astragalus fermentative matters medium and high dose groups were significantly increased(P<0.01),and the lev
