摘　　要：目的探讨肺炎支原体肺炎(MPP)患儿外周血白细胞自噬相关蛋白抗微管相关蛋白1轻链3(LC3)、Beclin1的表达及临床意义。方法采用前瞻性研究方法,收集2019年1月~2020年5月山西省人民医院住院的轻症、重症肺炎支原体肺炎患儿血液标本及正常门诊体检儿童外周血,分离白细胞,采用Western Blot检测白细胞中LC3II、LC3I、Beclin-1的表达量,计算LC3II/I比例;ELISA法检测血清白细胞介素-6(IL-6)、肿瘤坏死因子α(TNF-α)含量;比较上述指标在3组儿童血清中的表达量,并分析自噬和细胞因子之间的相关性。结果①共纳入MPP组50例,其中轻症28例,重症22例;对照组(正常体检儿童)25例。②MPP轻症组、重症组外周血白细胞中自噬相关蛋白LC3II/I比例及Beclin1表达量高于对照组,MPP重症组高于轻症组,差异有统计学意义(P<0.05)。③MPP轻症组、重症组IL-6、TNF-α水平高于对照组,MPP重症组IL-6、TNF-α水平高于轻症组,差异有统计学意义(P<0.05)。④自噬相关蛋白LC3II/I比例、Beclin1表达量与IL-6、TNF-α均成正相关(P<0.05)。结论MPP中发生了自噬,自噬和病情程度有关,病情越重,自噬程度越高。MPP中细胞因子水平增高,炎症反应明显。自噬和细胞因子呈正相关,提示MPP发生自噬影响细胞因子改变,从而影响疾病严重程度。Objective To study the expression and clinical significance of autophagy related protein anti-tubule related protein 1(microtubule-associated protein 1light chain 3,LC3)and Beclin 1 in peripheral blood of children with mycoplasma pneumoniae pneumonia(MPP).Methods In this prospective study,blood samples of children with mild and severe Mycoplasma pneumoniae pneumonia and peripheral blood of healthy children were collected from January 2019 to May 2020 in Shanxi Provincial People's Hospital.White blood cells were isolated.The expression levels of LC3II,LC3I and Beclin 1 in white blood cells were detected by Western blot,and the LC3II/I ratio was calculated.Serum interleukin 6(IL-6)and tumor necrosis factor-α(TNF-α)were detected by ELISA.The expression levels of the above indexes in the serum of the three groups were compared,and the correlation between autophagy and cytokines was analyzed.Results A total of 50 cases were included in the MPP group,including 28 cases of mild disease,22 cases of severe disease and 25 cases of control group.The proportion of autophagy related protein LC3II/I and the expression of Beclin1 in peripheral blood leukocytes of mild and severe MPP groups were higher than those of control group,and the expression of autophagy related protein LC3II/I in severe MPP group was higher than that of mild MPP group(P<0.05).The expression of IL-6 and TNF-6 in mild MPP group and severe MPP groupαThe levels of IL-6 and TN-6 in MPP group were higher than those in control groupαThe difference was statistically significant(P<0.05).Autophagy related protein LC3II/I ratio,Beclin1 expression and IL-6,TNF-αThere was a positive correlation between them(P<0.05).Conclusion Autophagy occurs in peripheral leukocytes in MPP,wihch is related to the degree of illness,the more serious the disease,the higher the degree of autophagy.The inflammatory response is obvious when Cytokines increases in MP.Autophagy and cytokines are positively correlated,suggesting that MP autophagy affects the changes of cytokine,thus aff
