摘  要:目的分析百草枯所致肺损伤大鼠肺组织的凋亡情况,并基于基因芯片技术探讨大鼠肺组织凋亡相关基因表达变化。方法取18只SD大鼠,随机分为百草枯组和阴性对照组各9只,给予百草枯组大鼠腹腔注射百草枯建立急性肺损伤模型,阴性对照组大鼠腹腔注射等量的生理盐水。应用脱氧核苷酸末端转移酶介导的dUTP缺口末端标记(TUNEL)法检测两组大鼠肺组织细胞凋亡情况。采用大鼠细胞凋亡PCR芯片检测两组大鼠肺组织凋亡相关基因的表达情况,并对测序数据进行生物信息学分析,筛选出有意义的差异表达基因进行实时定量PCR验证。结果百草枯组大鼠肺组织中TUNEL阳性细胞比例高于阴性对照组(P<0.05)。共筛选出3个差异表达基因,百草枯组大鼠肺组织的TNFSF10、FASLG和TNFRSF1B基因表达降低,与PCR验证结果具有一致性。结论百草枯导致的肺损伤大鼠肺组织存在凋亡,可能与凋亡相关基因TNFSF10、FASLG和TNFRSF1B的表达下调有关。Objective To analyze the apoptosis in lung tissues of rats with lung injury induced by paraquat,and to explore the changes in the expression of apoptosis-related genes in rat lung tissues by gene chip technology.Methods Eighteen SD rats were randomized to paraquat group(n=9)or negative control group(n=9),and intraperitoneal injection of paraquat was conducted in the paraquat group to establish a model of acute lung injury,whereas intraperitoneal injection of equivalent volume of normal saline was conducted in the negative control group.Terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling(TUNEL)assay was applied to detecting cell apoptosis in rat lung tissues of both groups.The apoptosis-related genes expression in rat lung tissues of both groups was determined by using PCR chip of rat cell apoptosis,and bioinformatics analysis was performed on the sequencing data,then real-time quantitative PCR was used to verify the significantly differentially-expressed genes.Results The ratio of TUNEL positive cells in rat lung tissues in the paraquat group was higher than that in the negative control group(P<0.05).Totally three differently expressed genes were screened,the expression of TNFSF10,FASLG and TNFRSF1B genes in rat lung tissues was decreased in the paraquat group,and the result was consistent with PCR verification result.Conclusion Apoptosis exists in lung tissues of rats with lung injury induced by paraquat,which may be related to down-regulation of the expression of apoptosis-related genes TNFSF10,FASLG and TNFRSF1B.
