摘 要:目的研究人参皂苷Rb1对雌性大鼠产后压力性尿失禁尿道周围组织损伤修复的作用,并探索其中机制。方法采用球囊扩张法模拟产后压力性尿失禁模型,将模型成功SD大鼠分为模型组、人参皂苷Rb1组。造模后第2天药物干预,人参皂苷Rb1组每日人参皂苷Rb11mg,模型组予等量0.9%氯化钠注射液进行灌胃,连续用药7 d。各组大鼠在给药前后分别行喷嚏实验,给药结束后用反转录-聚合酶链反应(RT-PCR)法检测各组大鼠尿道组织中转化生长因子(TGF)-β1 mRNA的表达含量。结果人参皂苷Rb1组治疗后喷嚏实验阳性率提升,与模型组比较差异有统计学意义(P<0.01)。Masson′s染色显示人参皂苷Rb1组尿道结构较模型组恢复好,RT-PCR结果表明模型组尿道组织中TGF-β1 mRNA的表达下降,与正常对照组、人参皂苷Rb1组比较差异有统计学意义(P<0.01)。结论人参皂苷Rb1治疗压力性尿失禁有效,其机制可能与提高尿道组织中TGF-β1的表达含量,促进损伤肌肉组织修复,促进胶原蛋白形成有关。Objective To determine the effect of ginsenoside Rg1 on the repair of periurethral tissue injury in female rats with postpartum stress urinary incontinence,and to investigate its potential mechanism.Methods The balloon expansion method was used to simulate the postpartum stress urinary incontinence model,and the SD rat models were successfully established.All the rats were randomly divided into the model group,study group and normal control group.The drug intervention was performed at 2d after modeling.The study group was given 1mg ginsenoside Rg1 daily,and the model group was given the same amount of normal saline byintragastric administration for 7 consecutive days.Sneeze test was performed in each group at baseline and after drug administration.After the administration,RT-PCR was used to examine the mRNA expression level of transforming growth factorβ1(TGF-β1)in the urethral tissue of rats in each group.Results The positive rate of sneezing test in the study group decreased after the treatment,and there was statistically significant difference compared with that in the model group(P<0.01).Masson′s staining showed that the mRNA expression level of TGF-β1 in the urethral tissue of the model group decreased,and there was statistically significant difference compared with that in the normal control group and the study group,respectively(both P<0.01).Conclusion Ginsenoside Rg1 is effective in the treatment of stress urinary incontinence,and its mechanism may be related to the increased expression level of TGF-β1 in urethral tissue,which promotes the repair of injured muscle tissue and the formation of collagen.
