摘  要:目的探讨Egr1-survivin shRNA转染联合放疗下调survivin表达对食管鳞癌KYSE150细胞凋亡及放射敏感性影响。方法将合成的辐射表达载体Egr1-survivin shRNA经Lipofectamine^(TM)2000转染KYSE150细胞后给予放疗,并以YM155联合放疗为主要对照,通过qPCR、Western-Blot检测处理后24 h各组细胞survivin的mRNA及蛋白表达,用流式细胞仪检测处理后24 h细胞凋亡率变化。结果 KYSE150细胞经Egr1-survivin shRNA质粒转染联合放疗处理后survivin mRNA及蛋白的表达量不但明显低于空白对照组和空载体对照组,也明显低于YM155处理组。经Egr1-survivin shRNA联合放疗处理后,KYSE150细胞的凋亡率明显增加,高于YM155联合放疗组,更明显高于空白对照组和空载体对照组,(P<0.01)。结论辐射表达载体Egr1-survivin shRNA联合放疗相比于YM155联合放疗可进一步增加食管鳞癌KYSE150细胞凋亡,增强其放射敏感性,是一种值得探索的辅助放疗途径。Objective To investigate the effects of transfected Egr1-survivin shRNA combined irradiation to down-regulate survivin protein on the apoptosis and radiosensitivity esophageal squamous cell carcinoma KYSE150 cells.Methods The radiative expression vector Egr1-survivin shRNA was designed to transfect into KYSE150 cells by LipofectamineTM2000,then radiotherapy been done.The expression level of survivin mRNA,proteins and the cell apoptosis of each treatment group was detected after treatments 24 h by qPCR,Westem Blot and flow cytometry respectively.Results In the KYSE150 cells,the expression levels of survivin mRNA and protein were significantly down-regulated in transfected Egr1-survivin shRNA plasmid combined irradiation group than that in YM155 combined irradiation group,the blank control group,and the blank vector control group.Compared with the YM155 combined irradiation group,the blank control group,and the blank vector control group,the apoptosis rate were clearly decreased in Egr1-survivin shRNA combined irradiation group(P<0.01 respectively).Conclusion The radiative expression vector Egr1-survivin shRNA combined irradiation can promote KYSE150 cells’apoptosis and enhance its radiotherapy sensitivity compared with the YM155 combined irradiation group.It is maybe a hopeful therapeutic method in conjunction with radiotherapy.
