摘 要:在现代细胞生物学研究中,为了揭示生物大分子空间定位、相互关系及其功能,高效快速地从体外培养的细胞中分离纯化出纯度较高的细胞器就显得至关重要。本综述主要介绍两种常见的分离纯化方式——离心分离和亲和免疫纯化分离,以及在细胞器分离纯化过程中,细胞破碎方法和溶剂的选择,两种方法的优缺点和纯化后的鉴定方法。In modern cell biology research,in order to reveal the spatial localization,interrelationship and function of biomacromolecules,it is very important to efficiently isolate and purify organelles with high purity from cultured cells in vitro.This review mainly introduces two common separation and purification methods:centrifugal separation and affinity immunopurification separation,as well as the selection of cell crushing methods and solvents in the process of organelle separation and purification,the advantages and disadvantages of the two methods and the identification methods after purification.
