摘  要:目的:研究扁蒴藤素下调整合素β1(integrinβ1)对肺癌A549细胞侵袭、迁移的影响。方法:体外培养肺癌A549细胞,完全培养液稀释后扁蒴藤素终浓度分别为0,5,10,20,40,80μg·ml^(-1)继续培养48 h,检测450 nm下各孔细胞光密度(OD)值,并计算半抑制浓度(I_(C50))。实验分为空白对照组、扁蒴藤素组、integrinβ1抑制剂(HMIβ1-1)组、扁蒴藤素+integrinβ1抑制剂组。空白对照组仅添加培养液,扁蒴藤素组添加终浓度为25.39μg·ml^(-1)扁蒴藤素的培养液,integrinβ1抑制剂组添加终浓度为10μg·ml^(-1)HMIβ1-1的培养液,扁蒴藤素+integrinβ1抑制剂组在扁蒴藤素组基础上添加10μg·ml^(-1)HMIβ1-1。Transwell检测细胞侵袭和迁移情况;划痕实验检测细胞迁移情况;实时荧光定量PCR(qRT-PCR)检测细胞中integrinβ1 mRNA表达情况;蛋白免疫印迹检测细胞中integrinβ1、纤维粘连蛋白(FN)、基质金属蛋白酶2(MMP2)蛋白表达情况。结果:与0和5μg·ml^(-1)扁蒴藤素相比,10,20,40,80μg·ml^(-1)扁蒴藤素OD值依次降低(P<0.05),I_(C50)为25.39μg·ml^(-1)。与空白对照组相比,扁蒴藤素组、integrinβ1抑制剂组侵袭、迁移细胞数量、划痕愈合率、细胞中integrinβ1 mRNA和蛋白水平、FN、MMP2蛋白水平显著降低(P<0.05);与扁蒴藤素组、integrinβ1抑制剂组相比,扁蒴藤素+integrinβ1抑制剂组侵袭、迁移细胞数量,划痕愈合率、细胞中integrinβ1 mRNA和蛋白水平、FN蛋白水平显著降低(P<0.05)。结论:扁蒴藤素可下调integrinβ1从而抑制肺癌A549细胞侵袭、迁移。Objective:To study the effects of pristimerin on the invasion and migration of A549 cells by down-regulating integrinβ1.Methods:Lung cancer A549 cells were cultured in vitro.The final concentration of pristimerin was 0,5,10,20,40 and 80μg·ml^(-1)after dilution of the complete culture medium.After 48 hours of continuous culture,the OD of each cell was detected at 450 nm,and the I_(C50)was calculated.The experiment was divided into blank control group,pristimerin group,integrinβ1 inhibitor(HMIβ1-1)group and pristimerin+integrinβ1 inhibitor group.The blank control group only added culture medium,pristimerin group added culture medium with pristimerin at final concentration of 25.39μg·ml^(-1),integrinβ1 inhibitor group added culture medium with HMIβ1-1 at final concentration of 10μg·ml^(-1),and pristimerin+integrinβ1 inhibitor group added 10μg·ml^(-1)HMIβ1-1 on the basis of pristimerin group.Transwell was used to detect cell invasion and migration;scratch test was used to detect cell migration;real-time fluorescence quantitative PCR(qRT-PCR)was used to detect the expression of integrinβ1 mRNA in cells;Western blot was used to detect the expressions of integrinβ1,fibronectin(FN)and matrix metalloproteinase 2(MMP2)protein in cells.Results:Compared with that of 0 and 5μg·ml^(-1)pristimerin,the OD of 10,20,40 and 80μg·ml^(-1)pristimerin decreased successively(P<0.05),and the I_(C50)was 25.39μg·ml^(-1).Compared with those in the blank control group,the number of invasion and migration cells,rate of scratch healing,levels of integrinβ1 mRNA and protein,FN and MMP2 protein decreased in pristimerin group and integrinβ1 inhibitor group(P<0.05);compared with those in pristimerin group and integrinβ1 inhibitor group,the number of cell invasion and migration,rate of scratch healing,levels of integrinβ1 mRNA and protein,FN protein decreased in pristimerin+integrinβ1 inhibitor group(P<0.05).Conclusion:Pristimerin can down-regulate integrinβ1 and inhibit the invasion and migration of A549 cells.
