摘 要:目的本研究旨在研究慢性鼻窦炎伴鼻息肉(chronic rhinosinusitis with nasal polyps,CRSwNP)患者中不同嗜酸性粒细胞(Eos)比例的鼻息肉黏膜上皮紧密连接(tight junctions,TJs)的表达情况。方法在首都医科大学附属北京同仁医院鼻过敏科手术患者中,收集5例正常对照鼻黏膜和17例CRSwNP鼻息肉组织,应用实时荧光定量PCR和免疫荧光共聚焦显微镜检测TJs mRNA和蛋白的表达情况。结果实时荧光定量PCR结果显示claudin-1、claudin-4、claudin-7、occludin、ZO-1和ZO-2 mRNA在CRSwNP组表达较正常对照组下调,并且呈现Eos越多,表达越少的趋势;免疫荧光共聚焦显微镜检测同样发现CRSwNP中claudin-1、claudin-4、claudin-7、occludin表达减弱与Eos浸润程度呈一定相关性。结论CRSwNP患者鼻息肉黏膜上皮细胞多种TJs表达下降,Eos浸润程度可能对TJs的表达下降具有一定的影响。OBJECTIVE To investigate the expression of tight junctions(TJs)on the epithelial cells of chronic rhinosinusitis with nasal polyps(CRSwNP)of different eosinophils(Eos)proportions.METHODSFive normal nasal mucosa samples and 17 CRSwNP mucosa samples of patients who had endonasal endoscopic surgery were enrolled.Real-time fluorescence quantitative PCR was conducted to evaluate the mRNA expression of claudin-1,claudin-4,claudin-7,occludin,ZO-1 and ZO-2 and immunofluorescence was performed to exam the expression of claudin-1,claudin-4,claudin-7,occludin.RESULTSThem RNA expressions of claudin-1,claudin-4,claudin-7,occludin,ZO-1 and ZO-2 were decreased in CRSwNP with the increasing of Eos proportions,and the immunofluorescence also demonstrated discontinuous and broken expression of claudin-1,claudin-4,claudin-7 and occludin in an Eos-related way.CONCLUSION Our data suggest that Eos proportions may influence epithelial barrier function in CRSwNP patients by regulating TJ protein expression.
