摘 要:目的利用气质联用法(GC-MS)测定石菖蒲挥发油化学成分的含量,观察石菖蒲挥发油对乳鼠心肌细胞活力的影响。方法采用水蒸气蒸馏法提取石菖蒲挥发油,用GC-MS分离鉴定其化学成分,并运用面积归一法计算各成分含量;正常乳鼠心肌细胞分为正常培养组、基质组(缺氧液处理细胞)和石菖蒲组(60、80、100、120、140、160、180及200 mg/L石菖蒲挥发油培养3~5 d),采用四氮唑蓝(MTT)比色法测定细胞活力。结果共检出29个化合物,占挥发油总量的95.78%,主要含有甲基乙丁香酚、甲基异丁香酚、异丁香粉甲醚、α-细辛醚、β-细辛醚等,其中β-细辛醚含量最高(54.75%)、其次为α-细辛醚(32.18%)、异丁香粉甲醚(2.27%)、甲基异丁香酚(1.73%)及甲基乙丁香酚(1.60%);基质组与正常培养组心肌细胞存活率比较差异无统计学意义(P>0.05);与基质组比较,60~180 mg/L石菖蒲挥发油组细胞存活率升高,200 mg/L石菖蒲挥发油组细胞存活率降低,差异有统计学意义(P<0.05)。结论石菖蒲挥发油中含量最高的是β-细辛醚,可提高乳鼠心肌细胞活力。Objective To investigate the chemical constituents of volatile oil in acorus gramineus by gas chromatography-mass spectroscopy(GC-MS)and to observe the effects of volatile oil on the activity of cardiomyocytes in newborn rats.Methods The volatile oil was extracted by steam distillation,separated,and identified by GC-MS;the content of each component was calculated by area normalization method.Cardiomyocytes were divided into culture group,stroma group(anoxic fluid treated cells)and acorus gramineus group(60,80,100,120,140,160,180,and 200 mg/L acorus gramineus volatile oil cultured for 3 to 5 days).Cell activity was measured by MTT colorimetry.Results Twenty-nine compounds were detected,accounting for 95.78%of the total volatile oil.The oil mainly contains methyl ethyl eugenol,methyl isoeugenol,isoclove powder methyl ether,α-asarone,β-asarone,etc.The content of β-asarone was the highest(54.75%),followed by 32.18%α-asarone,2.27%isoclove powder methyl ether,1.73%methyl isoeugenol,and 1.60%methyl ethyl eugenol.There was no significant difference in cardiomyocytes survival rate between the stroma group and culture group(P>0.05).Compared with the stroma group,the cell survival rate of the acorus gramineus volatile oil group at the concentration of 60-180 mg/L increased,and the cell survival rate of acorus gramineus volatile oil group at the concentration of 200 mg/L decreased,differences showed statistical significance(P<0.05).Conclusion β-asarone is the highest content in volatile oil of acorus tatarinowii and it can improve the activity of cardiomyocytes of newborn rats.
