摘  要:目的建立同时测定妇乐颗粒中绿原酸、马钱苷、咖啡酸、甘草苷、甘草酸铵、大黄酚、大黄素、芍药苷、川楝素和延胡索乙素含量的高效液相色谱(HPLC)法。方法色谱柱为Agilent TC-C_(18)(250 mm×4.6 mm,5μm),以甲醇(A)-0.1%磷酸溶液(B)为流动相,流速1.0 ml/min;检测波长:327 nm(绿原酸、咖啡酸),237 nm(芍药苷,马钱苷,甘草苷,甘草酸铵),254 nm(大黄素,大黄酚),280 nm(川楝素,延胡索乙素);柱温:30℃,进样量:10μl。结果绿原酸、马钱苷、咖啡酸、甘草苷、甘草酸铵、大黄酚、大黄素、芍药苷、川楝素和延胡索乙素的线性范围分别为5.58~55.75,1.32~13.20,1.01~10.05,2.74~27.44,1.56~15.56,1.52~15.17,1.81~18.08,4.48~44.81,0.55~5.53,4.10~41.02μg/ml(r≥0.9991);平均加样回收率为98.9%~101.9%(RSD<2.0%,n=6)。结论本法简便、准确、重复性好,可作为妇乐颗粒的质量控制方法。Objective To develop an HPLC method for simultaneous determination of chlorogenic acid,loganin,caffeic acid,liquiritin,ammonium glycyrrhizinate,chrysophanol,emodin,paeoniflorin,toosendanin and tetrahydropalmatine in Fule Granules.Methods The separation was carried out on an Agilent TC-C_(18)(250 mm×4.6 mm,5μm)column with the mobile phase consisted of methanol(A)-0.1%phosphoric acid(B)in a gradient mode at a flow rate of 1.0 ml/min.The detection wavelength was set at 327 nm for chlorogenic acid and caffeic acid;237 nm for paeoniflorin,loganin,liquiritin and ammonium glycyrrhizinate;254 nm for chrysophanol and emodin;280 nm for toosendanin and tetrahydropalmatine.The column temperature was set at 30℃and the injection volume was 10μl.Results Chlorogenic acid,loganin,caffeic acid,liquiritin,ammonium glycyrrhizinate,chrysophanol,emodin,paeoniflorin,toosendanin and tetrahydropalmatine showed good linar relationships within the ranges of 5.58-55.75,1.32-13.20,1.01-10.05,2.74-27.44,1.56-15.56,1.52-15.17,1.81-18.08,4.48-44.81,0.55-5.53 and 4.10-41.02μg/ml(r≥0.9991).The average recoveries were 98.9%-101.9%(RSDs<2.0%,n=6).Conclusion The method is simple,accurate and reproducible,and can be used for the quality control of Fule Granules.
