摘 要:为获知太岁中古菌准确的菌群结构及分析其与原有结果的不同,在太岁组织表里层不同位置重复取样,采用适合太岁材料特性的热溶解膜过滤法处理后提取DNA,随后利用Illumina MiSeq高通量测序平台测定古菌16S rDNA V3~V4区扩增序列。结果显示:供试太岁所含古菌分属16个门、27个纲、39个目、64个属,相对丰度大于0.01%的属有22个;表层的多样性高于里层,由外至内有物种消失现象;重复取样间存在大量非共有菌群;甲烷杆菌属(Methanobacterium)是有高丰度OTU支撑的优势菌群,相对丰度在43.8595%~96.8110%,是用常规方法提取DNA测知的优势属中,唯一仍保持优势地位的属,且在内部样品中的相对丰度较之前测定值分别提升39.5542%和65.9244%。上述结果表明,甲烷杆菌属是太岁所含古菌的优势菌群,在其形成中存在作用。To obtain the exact structure of archaeal flora in Taisui and compare it with the results got by conventional DNA extraction methods,samples were repeatedly extracted from both inner and outer layers of Taisui tissue,and DNAs were extracted from the samples after treatment with membrane filtration after heating dissolving under conditions suitable for Taisui tissue.The amplified sequences of the archaeal 16S rDNA V3~V4 region were determined on the Illumina MiSeq high-throughput sequencing platform.The result showed that the archaea in the tested Taisui included 16 phyla,27 classes,39 orders and 64 genera.There were 22 genera with relative abundance greater than 0.01%.Flora diversity in the outer layer was higher than that in the inner layer.From the outer layer to the inner,there was a tendency of species disappearance.Many non-common archaea were found among the repeatedly extracted samples.Methanobacterium was a dominant genus supported by high abundance OTU and its relative abundance ranged from 43.8595%to 96.8110%.It is the only genus that still maintains its dominant position among the dominant genera ob-tained by conventional DNA extraction methods,and the range of its relative abundance in the internal sam-ples increased by 39.5542%and 65.9244%,compared with that by conventional methods.The results demonstrated that Methanobacterium is the dominant genus of Taisui archaea,which plays a role in the for-mation of Taisui.
