摘 要:目的:探究软坚散结方对肾透明细胞癌(ccRCC)细胞(7860细胞系)裸鼠移植瘤生长抑制作用及对肿瘤组织Notch及凋亡信号通路相关基因蛋白的影响。方法:通过成功建立7860细胞裸鼠移植瘤模型,分为模型组、DAPT组(γ-secretase抑制剂)、软坚散结方组,分别给药4周后,检测各组瘤重,瘤体积;HE染色观察肿瘤细胞的形态变化;免疫组化检测肿瘤组织中Notch1、NICD、Bcl-2、Bax蛋白表达;RT-PCR检测Notch1、Bcl-2、Bax mRNA含量。结果:与模型组比较,软坚散结方组、DAPT组均可显著降低裸鼠体内肿瘤质量,缩小肿瘤体积,降低Notch1、Bcl-2 mRNA表达和Notch1、NICD、Bcl-2阳性率,升高Bax表达及阳性率(P<0.05);与DAPT组比较,软坚散结方组可降低裸鼠体内肿瘤质量、瘤体积、Bcl-2表达及NICD、Bcl-2性率,升高Bax表达及阳性率(P<0.05),差异有统计学意义。结论:软坚散结方可抑制7860细胞裸鼠移植瘤生长,其机制可能与靶向作用Notch信号通路从而诱导细胞凋亡相关。Objective:To investigate effects of Ruanjian Sanjie Decoction targeting Notch and apoptosis signal pathway on the growth of renal clear cell carcinoma(ccRCC)cell line(7860)transplanted tumor in nude mice.Methods:To establish the transplanted tumor model of 7860 cells in nude mice,and then divided into model group,DAPT group(Notch blocker)and Ruanjian Sanjie Decoction group.After successful modeling,the drugs was given for 4 weeks.The effect of Ruanjian Sanjie decoction on tumor weight and volume in nude mice were observed.The morphological changes of tumor cells were observed by HE staining,and Real-time PCR was used to detect the expressions of Notch1,Bcl-2 and Bax mRNA in tumor tissues.Results:Compared with the model group,Ruanjian Sanjie Decoction group and DAPT group could significantly reduce tumor mass and volume and decrease the expression of Notch1,Bcl-2 mRNA and the positive rate of Notch1,NICD and Bcl-2,meanwhile increase the expression of Bax and positive rate in nude mice(P<0.05).Compared with DAPT group,Ruanjian Sanjie decoction could decrease the tumor mass and volume,expression of Bcl-2 and positive rate of NICD,Bcl-2,and increase expression of Bax and positive rate in nude mice(P<0.05).Conclusion:Ruanjian Sanjie Decoction can inhibit the growth of transplanted tumor of 7860 cells in nude mice by targeting Notch and apoptosis signal pathway.
