摘 要:以青龙胆(Tinospora sagittata)的节间为研究对象,探索不同节间对腋芽诱导的影响,并针对增殖后丛生芽孱弱的情况,对组培苗壮苗培养基进行筛选。结果表明,节间位置对腋芽的诱导率和褐化率影响显著,随着节间位置靠下,诱导率逐渐下降,褐化率逐渐上升。靠近顶端的①号节间腋芽诱导率最高,为93.3%,显著高于其他位置的节间,褐化率最低,为13.3%,>0.5 cm的腋芽数量为6.7个。最佳的壮苗培养基为MS+10 mL/L EDTA,组培苗的平均苗高为4.1 cm,平均叶片数量为6.5片,叶色浓绿。Tinospora sagittate nodal segments in different positions were used as initial explants to explore the effect on axillary induc⁃tion.Media were also screened for strengthening of weak plantlets.The results showed that positions of nodal segments significantly in⁃fluenced the axillary induction and browning rate.With the positions of nodal segments from top to base of stems,the axillary induction rate decreased but browning rate increased gradually.The axillary induction rate of top no.1 nodal segment was 93.3%,which was sig⁃nificant higher than other positons of nodal segments.The browning rate of top no.1 nodal segment was 13.3%and the number of axil⁃lary buds(>0.5 cm)was 6.7.Maximum average length of shoots(4.1 cm)and average number of leaves(6.5)was recorded in the opti⁃mal strengthening media of MS+10 mL/L EDTA.
