摘  要:目的:研究苏木提取物对被动型Heymann肾炎模型小鼠的影响及对足细胞自噬反应的作用机制。方法:将实验动物随机分成5组,即正常对照组、Heymann肾炎模型组和低、中、高剂量苏木提取物组,给药21 d后检测各组小鼠24 h尿蛋白、血清白蛋白、肌酐等指标;光镜观察肾组织病理变化,免疫荧光染色法检测肾脏C5b-9沉积,蛋白印迹法(Western blot,WB)检测相关蛋白表达。结果:(1)与正常对照组相比,模型组24 h尿蛋白、血肌酐均不同程度升高,血清白蛋白不同程度降低(P<0.05);模型组光镜下可见肾组织明显分布不均匀,肾小球明显增大,系膜区显著增宽;模型组肾小球毛细血管袢C5b-9沉积明显增强;模型组肾组织P62表达明显降低(P<0.05),ATG5、LC3Ⅱ表达均明显升高(P<0.05)。(2)与模型组相比,苏木提取物各剂量组24 h尿蛋白、肌酐均不同程度降低,血清白蛋白不同程度升高(P<0.05);肾组织排列较不规范,部分肾小球出现增大;肾小球毛细血管袢C5b-9的沉积有所减少,肾组织P62表达明显升高,ATG5、LC3Ⅱ表达均明显降低(P<0.05)。结论:苏木提取物对Heymann肾炎小鼠具有肾保护作用,通过调控自噬进而保护和修复足细胞是其部分作用机制。Objective:To investigate the effect of Hematoxylon Extract on passive Heymann nephritis model mice and the mechanism of podocyte autophagy reaction.Methods:The experimental animals were randomly divided into 5 groups:normal control group,Heymann nephritis model group and low,medium,high-dose Hematoxylon Extract group.After 21 days of medication,24 h urinary protein,serum albumin,creatinine and other indexes were detected in each group.The pathological changes of renal tissues were observed by light microscopy.Immunofluorescence staining was used to detect the deposition of C5B-9 in the kidney.Western blot(WB)was used to detect the related proteins expressions.Results:(1)Compared with normal control group,24 h urinary protein and serum creatinine in model group were increased to varying degrees,while serum albumin was decreased to varying degrees(P<0.05).Compared with the normal group,the distribution of renal tissue in the model group was significantly uneven,the glomerulus was significantly enlarged,and the mesangial area was significantly widened.The deposition of C5b-9 in glomerular capillary loop was significantly enhanced in model group.The expression of P62 in renal tissue of model group was significantly decreased(P<0.05),while the expressions of Atg5 and LC3 were significantly increased(P<0.05).(2)Compared with model group,24 h urinary protein and creatinine in Hematoxylon Extract groups were decreased in different degrees,while serum albumin was increased in different degrees(P<0.05).the renal tissue arrangement was not standardized to varying degrees,and some glomeruli were enlarged.The deposition of glomerular capillary loop C5b-9 was reduced,the expression of P62 in renal tissue was significantly increased,and the expressions of ATG5 and LC3 were significantly decreased(P<0.05).Conclusion:Hema-toxylon Extract can protect the kidney of mice with Heymann nephritis,and the protection and repair of podocytes by regulating autophagy is part of its mechanism.
