猪多病原混合感染的实验室诊断

摘　　要：为对某养殖户发病猪只病因进行确诊,本试验采用常规PCR、荧光定量RT-PCR、细菌分离培养及鉴定、药敏试验等方法,结合流行病学调查、临床诊断及病理剖检对病猪进行诊断。病理剖检结果显示,病猪肝脏表面有出血点,肠系膜淋巴结肿大,肺肉质变,小肠、盲肠黏膜有溃疡;荧光定量RT-PCR方法检测结果显示,高致病性猪繁殖与呼吸综合征病毒(HP-PRRSV)和猪瘟病毒(CSFV)呈阳性;细菌分离培养鉴定及常规PCR方法确定有沙门菌感染。结果表明,发病猪为HP-PRRSV、CSFV和沙门菌的混合感染。根据确诊结果为该养殖户提出合理的防治措施建议,并对周边地区养殖场户提出预警及防治方法。In order to confirm the etiological factors of the diseased pigs in farm,conventional PCR,real-time RT-PCR,bacterial isolation and culture,biochemical identification and drug sensitivity test were used in this study,combined with epidemiological investigation,clinical diagnosis and pathologic autopsy to diagnose the sick pigs.The autopsy results showed that there were hemorrhagic spots on the liver surface,mesenteric lymph nodes enlargement,pulmonary cornification,ulceration on the intestinal and cecal mucosa of the sick pigs.The results of real-time RT-PCR showed that the highly pathogenic porcine reproductive and respiratory syndrome virus(HP-PRRSV)and classical swine fever virus(CSFV)were positive.Salmonella infection was identified by bacterial isolation,culture and comventional PCR.These data indicated that the cause of the disease was mixed infection of HP-PRRSV,CSFV and Salmonella.According to the confirmed results,reasonable prevention and control measures were proposed for the farmers,and early warning and prevention methods were proposed for farmers in the surrouding areas.
