摘 要:为了分析2016年广州市农贸市场所售的鸡肉中科瓦利斯沙门菌(Salmonella corvallis)耐药性及流行特征,参照国标检测方法(GB4789.4—2016),对广州市农贸市场采集的鸡肉样品进行沙门菌分离鉴定和血清学分型,用Kirby-Bauer法进行14种抗菌药物的耐药试验,用PCR方法检测携带的耐药基因,根据沙门菌脉冲场凝胶电泳(PFGE)分型试验方案进行分子分型。结果显示,21株科瓦利斯沙门菌中,对磺胺异噁唑耐药性最高(95.2%),其次为萘啶酸(90.5%)、四环素(66.7%),对环丙沙星的耐药率为19%,多重耐药菌检出率为81%。21株科瓦利斯沙门菌中喹诺酮类耐药决定区(QRDR)基因parC的突变率为100%,其第57位氨基酸苏氨酸突变为丝氨酸(Thr57→Ser),并且在质粒介导的喹诺酮类耐药(PMQR)基因中检出了qnrS、aac(6′)-Ib-cr和qnrB,检出率分别为90.5%、23.8%和9.5%。21株科瓦利斯沙门菌经PFGE分型后,结果显示其可分为7个血清型,其中具有1个优势克隆型(11株)。研究结果表明,广州市农贸市场鸡肉源科瓦利斯沙门菌分离株呈现耐药模式多样,多重耐药菌株严重,PFGE分子型别呈多态性,有同源菌株存在,提示存在共同来源或者交叉污染,应加强鸡肉中科瓦利斯沙门菌监测,同时为进一步做好食品中沙门菌的防控提供数据支持。In order to investigate the drug resistance and epidemiological characteristics of Salmonella corvallis isolated from chicken in Guangzhou city in 2016.Chicken samples were collected from the Guangzhou retail market,and Salmonella was isolated from the collected chicken samples and serologically typed according to the national standard(GB4789.4—2016).The Kirby-Bauer method was used to test the resistance of Salmonella to 14 antibiotics.The resistance genes were detected by PCR.Molecular typing was based on the Salmonella pulsed field gel electrophoresis(PFGE)typing.The results showed that among 21 strains of Salmonella corvallis,the highest resistance rate was sulfisoxazole(95.2%),followed by nalidixic acid(90.5%),tetracycline(66.7%),the resistance rate to ciprofloxacin was 19%and the detection rate of multidrug-resistant bacteria was 81%(resistant to three or more antibiotics).In 21 strains of Salmonella corvallis,the mutation of the quinolone resistance determining region(QRDR)gene parC was 100%,the 57th amino acid was mutated from threonine to serine(Thr57→Ser),and the detection rates of qnrS,aac(6′)-Ib-cr and qnrB were 90.5%,23.8%and 9.5%,respectively.7 genotypes were confirmed among 21 strains of Salmonella corvallis after PFGE typing,among which there was one dominant clonotype(11 strains).Our research showed that Salmonella covallis isolated from the chicken in Guangzhou have diverse drug resistance patterns,and multidrug-resistant strains are severe.PFGE molecular types are polymorphic,and there are homologous strains,suggesting the existence of a common source for cross-contamination.The result prompts us to strengthen surveillance of Salmonella corvallis in chicken and provide data support for further prevention and control of Salmonella in food.
