摘　　要：本试验旨在研究Toll样受体2(TLR2)在大鼠感染幽门螺杆菌(HP)前后及乙酸诱导的胃黏膜组织中的表达,探讨TLR2在HP感染致病机制中的作用。将30只大鼠随机分成3组,第1组为阴性对照组(正常饲养,不予处理),第2组为阳性对照组(采用乙酸诱导胃溃疡),第3组为HP感染组,分别连续7、14、21 d和30 d,建立胃溃疡模型,探讨TLR2在溃疡模型中的表达量。本试验采用qRT-PCR、免疫组化及Western Blot检测大鼠胃黏膜组织中TLR2的表达,苏木精-伊红(H.E.)染色法观察细胞形态及判断胃黏膜炎症程度,Giemsa染色检测胃黏膜组织中HP定植情况。结果显示:(1)TLR2在各组大鼠胃黏膜组织中均有表达;(2)TLR2在HP感染30 d组胃黏膜组织中表达高于其他处理组及正常组(P<0.001)。结果表明TLR2参与了HP致胃溃疡的发病过程,且在HP感染组中表达量呈抛物线变化。The purpose of this study was to study the expression of toll-like receptor 2(TLR2)in gastric mucosa of rats before and after Helicobacter pylori(HP)infection and acetic acid induced gastric ulcer,and to study the role of TLR2 in the pathogenesis of HP infection.30 rats were randomly divided into three groups:group 1 was negative control group(normal feeding,no treatment);group 2 was positive control group(gastric ulcer induced by acetic acid);group 3 was HP infection group.At 7,14,21 days and 30 days,a gastric ulcer model was established,and the expression of TLR2 in the ulcer model was explored.In this study,the methods of qRT-PCR,immunohistochemistry and Western Blot were used to detect the expression of TLR2 in gastric mucosa of rats.Hematoxylin-eosin(H.E.)staining was used to observe the morphology of cells and to judge the degree of inflammation of gastric mucosa,Giemsa staining was used to detect HP implantation in gastric mucosal tissue.The results showed that:(1)TLR2 was expressed in gastric mucosa of all groups of rats.(2)The expression of TLR2 in gastric mucosa of HP infection group was higher than that of other treatment group and normal group(P<0.001).The results indicate that TLR2 is involved in the pathogenesis of gastric ulcer induced by HP,and the expression level is parabola in HP infection group.
