摘 要:目的探究硬脂酰辅酶A去饱和酶(SCD)基因在宫颈癌患者中的表达、临床特征、预后及可能相关的作用机制。方法TCGA数据库下载宫颈癌病例临床及mRNA测序数据,对样本中SCD表达情况及临床数据进行统计分析,采用GEPIA数据库分析SCD对宫颈癌患者预后的影响,采用GSEA软件分析SCD基因在宫颈癌中可能的富集通路。结果TCGA数据库中,宫颈癌组织中SCD表达量明显高于正常宫颈组织(P=0.003)。TCGA数据库中306例宫颈癌患者数据分析显示,SCD在宫颈鳞状细胞癌中表达高于腺癌及其他类型肿瘤(P=0.001)。生存分析结果显示,SCD低表达组的预后明显优于SCD高表达组(log-rank P=0.0064,HR=1.9,P HR=0.0076)。GSEA分析显示,SCD基因高表达样本主要富集了基底转录因子通路、不饱和脂肪酸的合成信号通路、细胞分裂等。结论推测SCD基因在宫颈鳞癌组织中高表达,SCD是宫颈癌的不良预后因素,有望成为潜在的预后生物标志物和治疗靶点。Objective To identify the expression,clinical significance,prognosis and mechanism of SCD gene in cervical cancer.Methods The clinical and mRNA sequencing data of cervical cancer cases was download from TCGA database.The correlations between SCD expression and clinical data in the samples were analyzed.By GEPIA database analyzed the influence of SCD on the prognosis of cervical cancer patients,and by GSEA software analyzed the possible enrichment pathway of SCD gene in cervical cancer.Results The expression of SCD in cervical cancer was higher than that in normal cervical tissue(P=0.003).The data analysis of 306 cervical cancer patients in TCGA database indicated that the expression of SCD in cervical squamous cell carcinoma was higher than that in adenocarcinoma and other types of tumors(P=0.001).The survival analysis showed that the prognosis of SCD low expression group was significantly better than that of SCD high expression group on GEPIA databese(log-rank P=0.0064,HR=1.9,P HR=0.0076).GSEA analysis showed that the high expression samples of SCD gene mainly enriched in basal transcription factor pathway,unsaturated fatty acid synthesis signal pathway,cell division and so on.Conclusion SCD gene is highly expressed in cervical squamous cell carcinoma.Its high expression may be a potential prognostic biomarker and therapeutic target for cervical cancer.
