摘  要:以耐低温型克梅留斯基番茄为试材,采用农杆菌转化法将SlHSP17.7沉默载体成功转化到克梅留斯基番茄中,并进行相对电导率、丙二醛含量测定以及活性氧的NBT染色,研究沉默SlHSP17.7克梅留斯基番茄的耐低温性,以期明确SlHSP17.7与番茄耐低温性之间的关系。结果表明:沉默转基因株系膜脂过氧化程度和活性氧含量均高于对照,表现为不耐低温。可见,SlHSP17.7基因在提高番茄幼苗耐低温性方面发挥重要作用,这为提高番茄苗期耐低温能力及耐低温番茄新品种的培育提供新的思路。Chmielewskii which can resist low-temperature was used as the experiment material,SlHSP17.7 silencing vector was successfully transformed into chmielewskii by Agrobacterium tumefaciens-mediated method.In order to determine the relationship between SlHSP17.7 and low temperature tolerance of tomato,the low temperature tolerance of chmielewskii silencing SlHSP17.7 was studied by measuring the relative conductivity,malondialdehyde content and NBT staining of reactive oxygen species.The results showed that the degree of membrane lipid peroxidation and the content of reactive oxygen species of SlHSP17.7 silencing lines were higher than the control,and they were sensitive to low temperature.So,SlHSP17.7 plays an important role in improving the low temperature tolerance of tomato seedlings.The results provided a new idea for improving the low temperature tolerance of tomato seedlings and the cultivation of the resistant tomatoes.
