摘 要:以楸叶泡桐叶为底物,累积产氢量和产氢速率为主要考察目标,研究不同底物质量浓度对光合细菌HAU-M1同步糖化发酵产氢的影响,并对发酵过程中发酵液的pH值和还原糖浓度变化进行分析。实验结果表明:当底物浓度为25 g/L时,产氢效果最好,得到最大累积产氢量167.85 mL,氢气浓度达到39.5%;产氢高峰期集中在24~60 h,在36 h时,产氢速率达到最大值5.42 mL/h,在72 h后逐步停止产氢;反应液中的还原糖浓度在反应进行到0~12 h时快速上升,在12 h后逐渐下降,最终趋于平稳。利用修正的Gompertz方程对不同底物质量浓度下的产氢动力学进行分析,结果显示:当底物质量浓度为25 g/L时,光合产氢试验获得最大产氢潜能和产氢速率,分别为168.79 mL和6.51 mL/h。The effect of substrate concentration on photo-fermentation hydrogen production by HAU-M1 via simultaneous saccharification fermentation using Paulownia leaves as the substrate was studied.Taking cumulative hydrogen production and hydrogen production rate as assessment indicators,the photo-fermentation hydrogen production capabilities from leaves with different substrate concentrations are discussed.The variation of pH and reducing sugar concentration under different conditions is analyzed.It is found that the maximum cumulative hydrogen yield of 167.85 mL is obtained at substrate concentration 25 g/L,corresponding with the hydrogen content of 39.5%,and the maximum hydrogen production rate of 5.42 mL/h is obtained at 36 h.The hydrogen production cycle of Paulownia leaves is 72 hours,the peak period of hydrogen production is concentrated in 24-60 h.When the reaction proceeded from0-12 h.The reducing sugar concentration of the reaction solution increased rapidly,then gradually decreased after 12 h,and kept stable at last.The kinetics of hydrogen production under different substrates concentration are analyzed by using the modified Gompertz equation.The results showed that Paulownia leaves could be used as the substrate for photosynthetic hydrogen production.The maximum cumulative hydrogen yield and hydrogen production rate are achieved at substrate concentration 25 g/L,which are 168.79 mL and 6.51 mL/h,respectively.
