摘  要:采用L-苯丙氨酸和CuSO_(4)作为手性流动相添加剂,建立液相色谱法拆分安妥沙星对映体的方法。考察了手性添加剂的种类、浓度及流动相pH等对安妥沙星对映体分离的影响。采用Welch Ultimate XBC_(18)色谱柱(4.6 mm×250 mm,5μm),优化流动相为8 mmol/L L-苯丙氨酸溶液(含4 mmol/L的CuSO_(4),pH 3.5)-甲醇(80∶20,V∶V),流速:1.0 mL/min,检测波长:302 nm,结果左、右旋安妥沙星峰达到基线分离,分离度为6.6。A method of HPLC was established for separating enantiomers of antofloxacin by using Lphenylalanine and copper sulfate as chiral mobile phase additives.The factors for chiral separation of antofloxacin,including the type of chiral additive,concentration of chiral additive and pH of mobile phase,were investigated.A Welch Ultimate XB-C_(18)column(4.6 mm×250 mm,5μm)was used,and the optimized mobile phase was 8 mmol/L L-phenylalanine solution(containing 4 mmol/L copper sulfate,pH 3.5)-methanol(80∶20,V∶V),flow rate was 1.0 m L/min and detection wavelength was 302 nm.The enantiomers of antofloxacin were completely separated with a resolution of 6.6.
