摘 要:目的观察急性汞中毒对小鼠肝的损伤,并探讨内质网应激是否介导此损伤。方法选取8周龄健康雄性C57B/L6小鼠,随机分为对照组、HgCl_(2)染毒组、4-PBA预处理+HgCl_(2)染毒组,建模成功后,眼球取血检测小鼠肝功能变化;HE染色观察肝形态学变化;免疫组织化学染色及Western blot检测肝组织中内质网应激特异性蛋白Bip与CHOP的表达情况。结果与对照组相比,HgCl_(2)染毒组小鼠肝损伤明显,且Bip与CHOP表达显著增高,而4-PBA可下调Bip与CHOP表达水平从而减轻肝损伤。结论急性汞中毒可能通过内质网应激引起肝损伤。Objective To observe the liver injury caused by acute mercury poisoning and to explore whether it was mediated by endoplasmic reticulum stress.Methods Healthy male C57B/L6 mice aged 8 weeks were randomly divided into the control group,the HgCl_(2) group,and the 4-PBA pretreated+HgCl_(2) group.After successfully establishing mice models,liver function in mice was evaluated by testing eyeball blood.HE staining was applied to observe the morphological changes of liver tissues.Immunohistochemistry and Western Blot were used to detect the expression of endoplasmic reticulum stress-specific proteins Bip and CHOP.Results Compared with the control group,livers in mice poisoned with HgCl_(2) were significantly damaged,and the expression of Bip and CHOP in their liver tissues was significantly increased,while 4-PBA could down-regulate the expression of Bip and CHOP,thereby attenuate liver damage.Conclusion Acute mercury poisoning may cause liver damage through endoplasmic reticulum stress.
