摘  要:目的:研究miR-216a调控JAK2/STAT3通路对脓毒症诱导的急性呼吸窘迫综合征(ARDS)的作用机制。方法:60只SD大鼠,随机分为对照组和ARDS组,每组各30只,各组每天灌胃1次,持续7 d。末次灌胃后1 h在尾静脉处注射0.9%氯化钠液或内毒素造模。造模后检测各组呼吸、血气等指标,比较两组大鼠呼吸频率、氧分压指标;观测两组病理学变化;采用瞬时转染的方法检测肺泡巨噬细胞中的miR-216a表达量水平;采用western blot法探究miR-216a对肺泡巨噬细胞中JAK2/STAT3通路的影响。结果:相比对照组,ARDS组大鼠呼吸频率显著加快,氧分压明显降低,病理评分明显更高;转染后,肺部巨噬细胞内的miR-216a水平明显增加,相比于阴性对照组转染后的miR-216a表达量增加30-250倍,差异性具有统计学意义(P<0.05)。miR-216a肺部巨噬细胞中MMP2蛋白、p-JAK2蛋白、p-STAT蛋白的表达量均高于对照组,差异性有统计学意义(P<0.05)。结论:miR-216a对JAK2/STAT3通路的作用是正向调控促进脓毒症诱导的ARDS的发展进程。Objective:to study the mechanism of Mir-216 a regulating JAK2/STAT3 pathway on acute respiratory distress syndrome(ARDS)induced by sepsis.Methods:60 SD rats were randomly divided into control group and ARDS group(130 rats in each group).The MODEL was established by injecting normal saline or endotoxin into caudal vein 1 hour after the last gastric perfusion.After modeling,the respiratory and blood gas indexes of each group were measured,and the respiratory rate and oxygen partial pressure indexes of the two groups were compared;the pathological changes of the two groups were observed;the expression level of miR-216 a in alveolar macrophages was detected by transient transfection;the effect of miR-216 a on the JAK2/STAT3 pathway in alveolar macrophages was investigated by Western blot.Results:Compared with the control group,rats in the ARDS group have a significantly faster respiratory rate,a significantly lower oxygen partial pressure,and a significantly higher pathological score;after transfection,the level of miR-216 a in the lung macrophages increased significantly,compared to the negative control group The expression of miR-216 a after transfection increased by 30-250 times,and the difference was statistically significant(P<0.05).The expression levels of MMP2 protein,p-JAK2 protein and p-STAT protein in miR-216 a lung macrophages were higher than those of the control group,and the difference was statistically significant(P<0.05).Conclusions:Mir-216 a has positive effect on JAK2/STAT3 pathway.
