摘  要:目的采用网络药理学方法探讨淫羊藿苷治疗支气管哮喘可能的作用机制。方法基于中药系统药理学数据库与分析平台(TCMSP)获取淫羊藿苷的ADME相关参数。通过Pharm Mapper数据库预测淫羊藿苷的潜在靶标,Genecards数据库获取支气管哮喘相关靶点,将淫羊藿苷靶点与支气管哮喘靶点映射得到淫羊藿苷作用于支气管哮喘的潜在靶点。采用STRING在线数据库获取蛋白相互作用(PPI)数据并导入Cytoscape中构建PPI网络。利用Bioconductor生物信息软件包对作用靶点进行基因本体(GO)功能富集分析和京都基因与基因组(KEGG)数据库通路富集分析。结果通过筛选获得125个淫羊藿苷作用于支气管哮喘的潜在靶点。富集分析结果表明,淫羊藿苷主要涉及蛋白质丝氨酸/苏氨酸激酶活性、细胞因子受体结合、核受体活性、激活转录因子结合等生物过程,通过调节T细胞受体信号通路、Fox O信号通路、Relaxin信号通路和VEGF信号通路等通路从而发挥治疗支气管哮喘的作用。结论淫羊藿苷通过多个靶点、多条通路对支气管哮喘所引发的炎症反应、气道高反应性及气道重塑发挥治疗作用。Objective:To explore the possible mechanism of icariin in the treatment of bronchial asthma by network pharmacology.Methods:The ADME related parameters of icariin were obtained based on TCMSP.PharmMapper was used to predict potential targets of icariin.GeneCards was used to screen the targets of bronchial asthma.The potential targets of icariin on bronchial asthma were obtained by mapping icariin targets and bronchial asthma targets.STRING online database was used to obtain protein interaction(PPI)data and then import the data into Cytoscape to construct PPI network.Bioconductor was used to analyze the GO function and KEGG pathway enrichment of the targets.Results:A total of 125 potential targets of icariin on bronchial asthma were obtained.The enrichment analysis results showed that protein serine/threonine kinase activity,cytokine receptor binding,nuclear receptor activity,activate transcription factor binding and other biological processes were mainly involved by adjusting T cell receptor signaling pathway,FoxO signaling pathway,Relaxin signaling pathway,VEGF signaling pathway and other pathways to exert its treatment of bronchial asthma.Conclusion:Icariin exerts a therapeutic effect on the inflammatory response,airway hyperresponsiveness and airway remodeling caused by bronchial asthma through multiple targets and multiple pathways.
