扶正祛邪复方对人白血病细胞HL-60的增殖及凋亡的影响

摘　　要：目的研究扶正祛邪复方对人白血病细胞HL-60增殖及凋亡的影响及其作用机制。方法采用不同质量浓度扶正祛邪复方提取物体外干预HL-60细胞,台盼蓝染色计数法、CCK-8检测法检测白血病细胞抑制率,应用流式细胞术检测细胞周期及凋亡率。结果不同质量浓度的扶正祛邪复方作用于HL-60细胞均能抑制细胞增长,并且随着质量浓度的增长呈递增趋势,相同浓度组的细胞在24、48、72 h抑制率也有一定的时间依赖性。流式细胞术检测白血病细胞的细胞周期及凋亡率,与正常对照组相比,扶正祛邪复方各质量浓度组对HL-60细胞增殖抑制率及凋亡率均有明显作用,且有一定的质量浓度依赖性。结论扶正祛邪复方能抑制人白血病细胞HL-60的增殖,其作用机制可能与影响细胞周期及诱导细胞凋亡相关。Objective:To investigate the effect and mechanism of Fuzheng Quxie Compound on the proliferation and apoptosis of HL-60 cells in human leukemia cell.Methods:HL-60 cells were interfered by different concentrations of Chinese herbal compound in vitro.Trypan-blue staining and CCK-8 assay were used to detect the proliferation inhibition rate of leukemia cells,and flow cytometry was used to detect the cell cycle and apoptosis rate.Results:Different concentrations of Fuzheng Quxie Compound could inhibit the growth of HL-60 cells,and with the increase of concentration,the trend was increasing.In the same concentration group,the inhibition rates of 24 h,48 h and 72 h were also time-dependent.The cell cycle and apoptosis rate of leukemia cells were detected by flow cytometry.Compared with the normal control group,the inhibition rate and apoptosis rate of HL-60 cells in each concentration group of Fuzheng Quxie Compound were significantly increased,and there was a certain concentration dependence.Conclusion:Fuzheng Quxie Compound can inhibit the proliferation of HL-60 cells,whose mechanism may be related to the influence of cell cycle and inducing apoptosis.
