摘 要:目的观察补阳还五汤对脑出血大鼠脑组织内自噬相关蛋白缺氧诱导因子(HIF)-1α、基质细胞衍生因子(SDF)-1、趋化因子受体(CXCR)-4表达的影响。方法72只雄性SD大鼠随机等分为假手术组、脑出血模型组、补阳还五汤组和银杏叶片组。采用Rosenberg法建立脑出血大鼠模型,补阳还五汤26.0 g/(kg·d)灌胃干预,连续给药14 d。Western印迹法检测Beclin-1蛋白的表达,免疫组化法检测HIF-1α、CXCR-4和SDF-1α蛋白的表达,TUNEL法观察神经细胞凋亡情况。结果与模型组比较,补阳还五汤组脑组织神经细胞凋亡数量明显减少,HIF-1α、Beclin-1蛋白表达减少,SDF-1α、CXCR-4蛋白表达显著增加。结论补阳还五汤能够降低HIF-1α表达,进而通过调控SDF-1α/CXCR4信号传导通路抑制脑出血诱发的神经细胞凋亡。Objective Effect of Buyang Huanwu decoction on the expressions of HIF-1α,SDF-1,CXCR-4 in cerebral tissue of rats with cerebral hemorrhage.Methods According to Rosenberg method,cerebral hemorrhage rat model was replicated and Buyang Huanwu decoction was gavaged with 26.0 g/(kg·d)administration.Western blot was used for the detection of Beclin-1 protein,immunohistochemistry method was used for the detection of HIF-1α,SDF-1,CXCR-4 protein.Apoptosis was detected by TUNEL method.Results Compared with model group,the number of neuronal apoptosis was significantly decreased,expressions of HIF-1α,Beclin-1 were decreased,SDF-1,CXCR-4 protein were significantly increased in Buyang Huanwu decoction group.Conclusions Buyang Huanwu decoction could de crease HIF-1αexpression and regulate SDF-1α/CXCR4 autophagy the SDF-1α/CXCR4 Signal transduction pathway,then inhibit neuron apoptosis induced by cerebral hemorrhage.
