摘  要:目的建立HPLC-FLD法测定林蛙卵和林蛙油中雌二醇的含量。方法采用Extend C_(18)(250 mm×4.6 mm,5 μm)色谱柱;流动相为甲醇-水(55∶45,V/V);流速为1.0 mL·min^(-1);柱温为40℃;荧光检测器激发波长为282 nm,发射波长为315 nm;进样量为20 μL。结果雌二醇质量浓度在0.22~21.68 μg·mL^(-1)内与峰面积呈良好的线性关系(r=0.9999);精密度、重复性和稳定性均良好(RSD<2.0%),平均加样回收率为100.4%(RSD为1.4%)。结论该方法操作简单、准确、易重复,可用于林蛙卵和林蛙油中雌二醇的含量测定。Objective To determine the content of estradiol in eggs and oil of Rana Dybowskii by HPLC-FLD.Methods The Extend C18(250 mm×4.6 mm,5μm)column was used with mobile phase consisting of methanol-water(55:45,V/V).The flow rate was 1.0 mL·min^(-1) and the column temperature was 40℃.The fluorescence detector excitation wavelength was 282 nm,and the emission wavelength was 315 nm.The injection volume was 20μL.Results There was a good linearity between the estradiol concentration and the peak area at 0.22-21.68μg·mL^(-1)(r=0.9999).The precision test,repeatability test and stability test were all good(RSD<2.0%),and the average recovery was 100.4%(RSD=1.4%).Conclusion This method is simple,accurate and repeatable,and can be used to determine the content of estradiol in eggs and oil of Rana Dybowskii.
