石竹和瞿麦组织培养与试管开花技术研究

摘　　要：以石竹的种子和重瓣瞿麦的茎段为试材,采用植物组织培养技术,以MS为基本培养基,添加不同浓度的植物生长调节物质,经过种子萌发和无菌苗的增殖,腋芽的诱导及增殖,研究了石竹和瞿麦组织培养和试管开花的最佳培养组合以及不同因素对石竹和瞿麦试管苗成花的影响。结果表明:在MS+6-BA 0.5 mg/L+NAA 0.1 mg/L+蔗糖20 g/L+琼脂7.0 g/L,增殖培养28 d处理下,石竹平均苗高可达到4.2 cm,增殖系数可达到2.8倍;瞿麦增殖系数可达7.28倍;在MS+6-BA 0.5 mg/L+NAA 0.1 mg/L+多效唑40 mg/L+蔗糖40 g/L+琼脂7.0 g/L处理下,石竹平均高度为4.33,茎粗可达0.744cm;瞿麦在多种配方的培养基中均可开花,石竹始终没有开花。The seeds of Dianthus chinensis and the stems of Dianthus superbus‘Chongban’were used as explants to study the best combination of tissue culture and in vitro flowering of these two species,with MS was used as the basic medium supplied with different concentrations of plant growth regulators.The results showed that the most suitable formula for the proliferation of the two species was MS+6-BA 0.5 mg/L+NAA 0.1 mg/L+Sucrose 20 g/L+Agar 7.0 g/L,and the average seedling height of D.chinensis was 4.2 cm,the proliferation coefficient reached 2.8 times after 28 days’growth;while the proliferation coefficient of D.superbus can reached 7.28 times with 28 days’culture.The formula for strong seedlings culture is MS+6-BA 0.5 mg/L+NAA 0.1 mg/L+Paclobutrazol 40 mg/L+Sucrose 40 g/L+Agar 7.0 g/L.D.superbus can bloom in a variety of medium formulations,but never D.superbus blooms.
