摘　　要：目的研究三七皂苷R1(NR1)对血管紧张素Ⅱ(AngⅡ)诱导的小鼠主动脉血管平滑肌(MOVAS)细胞增殖、迁移的影响。方法将MOVAS细胞分为Control组、NR1组、AngⅡ组、AngⅡ+NR1组。采用细胞计数试剂盒-8(CCK-8)检测MOVAS细胞的增殖活性,采用Transwell小室和划痕愈合实验检测MOVAS细胞纵、横向迁移能力。观察NR1对AngⅡ诱导MOVAS细胞增殖、迁移的影响。结果MOVAS细胞经AngⅡ(5μM)作用后,其增殖活性、纵、横向迁移能力明显增加,与Control组相比差异有统计学意义(P<0.01);而加NR1(50μM)干预后,AngⅡ+NR1组细胞增殖活性降低、纵、横向迁移能力均下降,与AngⅡ组相比,差异有统计学意义(P<0.05或P<0.01)。结论NR1可抑制AngⅡ诱导的血管平滑肌细胞增殖、迁移,这可能是NR1预防和治疗动脉粥样硬化及血管再狭窄等血管增生性疾病的作用机制之一。Objective To investigate the effects of notoginseno-side R1(NR1)on proliferation and migration of mouse aortic vascular smooth muscle(MOVAS)cells induced by angiotensinⅡ(AngⅡ).Methods MOVAS cells were divided into control group,NR1 group,AngⅡgroup and AngⅡ+NR1 group.Cell counting kit-8(CCK-8)was used to evaluate cell proliferation activity.Transwell chamber and wound healing test were used to analyze cell vertical and horizontal migration capability.All methods were used to observe the effect of NR1 on the proliferation and migration of MOVAS cells induced by AngⅡ.Results Compared with the control group,the proliferation activity,vertical and horizontal migration ability of MOVAS cells treated with AngⅡ(5μm)were significantly increased(P<0.01).While the proliferation activity,vertical and horizontal migration ability of MOVAS cells treated with AngⅡ+NR1(50μm)were significantly decreased compared with the AngⅡgroup(P<0.05 or P<0.01).Conclusion NR1 exerts inhibitory effects on the proliferation and migration of vascular smooth muscle cells induced by AngⅡ,which may be one of the mechanisms that NR1 can prevent and treat the vascular proliferative diseases such as vascular restenosis and atherosclerosis.
