摘 要:目的:运用网络药理学方法探讨水陆二仙丹治疗肾病综合征的药理机制。方法:通过中药系统药理学数据库及分析平台、Swiss Target Prediction数据库分别获取水陆二仙丹药物主要活性成分和作用的靶点,在Genecard数据库中查找NS对应的靶点,将最终筛选的靶点建立靶点相互作用网络,并用DAVID数据库对靶点进行GO分析和KEGG通路分析,最终建立药物-成分-靶点-通路网络。结果:筛选得到水陆二仙丹16个活性成分,涉及86个作用靶点,靶点主要通过调控PI3K-AKT、RAS、Rap1、HIF-1、VEGF等信号通路来发挥治疗NS的作用。结论:黄酮类可能是水陆二仙丹治疗NS的物质基础,通过调节PI3K-AKT、RAS等通路从而修复受损的足细胞、降低肾小球灌注压等作用减轻NS蛋白尿。Objective:To investigate the pharmacological mechanism of shuiluerxiandan in the treatment of nephrotic syndrome by network pharmacology.Methods:Through TCMSP,Swiss Target Prediction 1 database access to shuiluerxiandan of main active ingredients and drug targets,in the Genecards database lookup NS corresponding targets,will eventually screening targets set targets for interaction network,the DAVID database was used for GO analysis and KEGG pathway analysis,eventually establish drug-composition-targets-pathways network.Results:Sixteen active components of shuiluerxiandan were screened,involving 86 targets.The targets mainly play the role of NS treatment by regulating the signaling pathways such as PI3K-AKT、RAS、Rap1、HIF-1、VEGF.Conclusion:Flavonoids may be the material basis for the treatment of NS by shuiluerxiandan.By regulating PI3K-AKT,RAS and other pathways,it can repair damaged podocytes and reduce glomerular perfusion pressure to reduce NS proteinuria.
