摘  要:目的探讨维生素C和多西环素(Doxycycline)联用对人肝癌细胞株HepG2细胞增殖的抑制作用及其作用机制。方法在体外用细胞培养技术培养人肝癌HepG2细胞,用维生素C和多西环素联合干预,倒置显微镜下观察肿瘤细胞的生长情况,MTT法检测Vitmin C和Doxycycline联合应用对HepG2细胞增殖活性的影响,同时测定肝癌细胞肿瘤标志物(AFP、γ-GT)和凋亡蛋白(Fas、Bcl-2、Bax)的含量。结果用维生素C和多西环素联合干预HepG2细胞72h,能明显抑制肿瘤细胞的增殖(p<0.05);肿瘤标志物(AFP、γ-GT)和凋亡抑制蛋白Bcl-2含量下降(p<0.05);凋亡蛋白(Fas、Bax)的含量升高(p<0.05)。结论维生素C和多西环素联合应用在肝癌HepG2细胞体外培养时能够抑制肿瘤细胞的生长,其作用机制与增加癌细胞的凋亡作用有关。Objective To investigate the inhibitory effect of vitamin C and doxycycline on the proliferation of human Hepatoma cell HepG2 and its mechanism.Method Human hepatoma HepG2 cells were cultured in vitro with cell culture technology,and combined intervention of vitamin C and doxycycline.The growth of tumor cells was observed under an inverted microscope.The MTT method was used to detect the effect of the combination of vitmin C and Doxycycline on the proliferation of HepG2 cells.The contents of tumor markers(AFP,γ-GT)and apoptosis proteins(Fas,Bcl-2,Bax)of liver cancer cells were measured.Result The combined intervention of vitamin C and doxycycline in HepG2 cells for 72 hours could significantly inhibit the proliferation of tumor cells(p<0.05);Tumor markers(AFP,γ-GT)and apoptotic inhibited proteins of Bcl-2 content decreased(p<0.05);the content of apoptotic proteins(Fas,Bax)increased(p<0.05).Conclusion The combined application of vitamin C and doxycycline could inhibit the growth of tumor cells when hepatocarcinoma HepG2 cells were cultured in vitro,and its mechanism of action was related to the increase in apoptosis of cancer cells.
