摘 要:建立四川牡丹皮的超高效液相色谱(UPLC)指纹图谱和对照指纹图谱,并同时测定样品中的4种成分的含量,为了解四川牡丹皮化学成分特征提供方法和依据。采用Ultimate UPLC LP-C18(2.1 mm×100 mm,1.8μm)色谱柱,以0.1%(体积分数)甲酸水溶液(A)-乙腈(B)为流动相,梯度洗脱,流量0.2 mL/min,柱温30℃,检测波长254 nm,进样量2μL。建立四川牡丹皮的UPLC指纹图谱并标定共有峰18个,对其中4个色谱峰进行指认。17个样品指纹图谱相似度在0.765~0.989之间,丹皮酚、芍药苷、丹皮酚原苷和苯甲酰芍苷4种成分含量分别在6.97~24.0 mg/g、14.8~36.9mg/g、4.05~73.5 mg/g、4.83~18.0 mg/g之间。2个市售安徽产牡丹皮指纹图谱与四川牡丹皮相比,其相似度低,四川牡丹皮成分数量和含量均更高。UPLC指纹图谱结合多成分含量测定能够反映四川牡丹皮化学成分特征,方法重现性好、简便可行、快速准确,可为四川牡丹皮和其他牡丹皮化学成分特征差异提供研究方法。To establish the UPLC fingerprint and the method of multicomponent detection of Paeonia decomposita Cortex for provide method and data basis to understanding the chemical constituents of Paeonia decomposita Cortex.UPLC analysis was performed on a Ultimate UPLC LP-C18 column(2.1 mm×100 mm,1.8μm).The mobile phase consisted 0.1%formic acid aqueous solution(A)and acetonitrile(B)with gradient elution.The flow rate was 0.2 mL/min,the detection wavelength was 254 nm,the column temperature was 30℃and the injection volume was 2μL.The fingerprint of Paeonia decomposita Cortex was established.There were 18 common peaks in the fingerprint and four of them were identified.The similarity values of the chromatographic fingerprint of 17 samples ranged from 0.765 to 0.989.Meanwhile,the content of four major chemical constituents,namely Paeonol,paeoniflorin,paeonolidie and benzoyl paeoniflorin were determined in the range of 6.97-24.0 mg/g,14.8-36.9 mg/g,4.05-73.5 mg/g,4.83-18.0 mg/g.Compared with two marketed Cortex Moutan from Anhui,the UPLC fingerprint similarity was lower and the number of components and its quantity of Paeonia decomposita Cortex were higher than those of Anhui Cortex Moutan.The chemical composition characteristics of Paeonia decomposita Cortex can be reflected by UPLC fingerprint and simultaneous determination of multiple components.The established method is accurate and sensitive,stable,and reproducible,and which could provide reference for study on difference of chemical composition between Paeonia decomposita Cortex and other Cortex Moutan.
