连云港地区甲型H1N1流感病毒HA1基因病原学特征研究

摘  要:目的对连云港地区2018年—2019年甲型H1N1流感病毒表面血凝素HA1基因特征及抗原变异情况进行分析。方法对2018年—2019年连云港地区流感监测样本进行毒株分离,将分离到的6株甲型H1N1流感病毒株,用RT-PCR法对HA1基因进行扩增并测序,运用Lasergene软件包中MegAlign软件进行核苷酸以及氨基酸分析,用MEGA 6.0软件进行流感序列进化分析,并进行在线糖基化分析。结果6株连云港流感分离株与推荐疫苗株的HA1基因相比,核苷酸同源性为98.5%~99.9%,氨基酸同源性为98.9%~100%。基因树分析,6株新分离的流感病毒HA1基因分属不同的进化亚簇。进行氨基酸分析,6株HA1氨基酸序列均发生Sa区181(S→T),在Sb区202(S→T),在Cb区91(S→R)变异,1株在Sb区204(D→V)发生变异,2株在Ca2区155(H→Y)发生变异,但受体结合位点和糖基化位点的氨基酸均未发生。结论2018年—2019年在连云港地区人群中流行的甲型H1N1流感病毒与推荐疫苗株相比,基因特征发生一定改变,需要进一步加强流感监测。Objective To study the molecular evolutional characteristics of the HA1 gene of the influenza A H1N1 virus isolated in Lianyungang during 2018-2019.Methods Influenza like illness(ILI)samples collected from sentinel hospitals were subjected to nucleic acid analysis.Six strains of influenza A H1N1 virus isolated in Lianyungang during 2018-2019 were selected.The HA1 gene sequence was amplified by RT-PCR and the sequence characteristics and changes were analyzed by Lasergene software.MEGA6.0 software was used for sequence alignment and phylogenetics analysis.Results Six strains of the influenza A H1N1 virus shared high nucleic acid sequence similarity in HA genes(98.5%-99.9%),and high amino acid sequence similarity(98.9%-100%).The phylogenetic analysis showed that the HA gene of 6 isolated strains were divided into different evolutionary lineages.Except for A/Michigan/45/2015(H1N1),6 isolates had a mutation of 181(S→T)at Sa antigenic site in HA1 domain,202(S→T)at Sb antigenic site,91(S→R)at Cb antigenic site.One had the mutation of 204(D→V)at Sb antigenic site.Two isolates had 155(H→Y)at Ca2 antigenic site.Sequence analysis indicated there was no mutation in the receptor binding sites and glycosylation sites.Conclusion In 2018—2019,the HA1 gene of the influenza A H1N1 virus prevalent in Lianyungang have changes in genetic characteristics compared with international vaccine strains.Further etiological should be enhanced.
