摘　　要：目的建立高效液相色谱法测定睫毛增长液化妆品中比马前列素的方法。方法样品经含50%乙腈的水溶液超声提取,加入NaCl和MgSO4沉淀,离心,上层有机相经微孔滤膜过滤后,以Agilent Poroshell 120 EC-C18(4.6 mm×100 mm,2.7μm)液相色谱柱进行分离,乙腈和水(30:70,V:V)等度洗脱,流速为0.5 ml/min,检测波长为210 nm,经外标法定量。疑似阳性样品采用高效液相色谱-串联质谱法进行确证。结果比马前列素的线性范围为0.5μg/ml~150μg/ml,相关系数(r)值为0.99996;检测限为1 mg/kg,不同基质比马前列素的加样回收率(n=6)为94.2%~103.5%,RSD为1.4%~3.0%。测定了10个不同厂家睫毛增长液化妆品样品,3批样品检出比马前列素,含量为0.026%~0.030%。结论该方法可用于睫毛增长液化妆品中比马前列素的测定。Objective A method using high performance liquid chromatography was developed and validated for the determination of bimatoprost in eyelash enhancing cosmetics.Methods The cosmetic samples were supersonically extracted with 50%acetonitrile water solution and then a salt mixture(NaCl,MgS04) was added to the solution to induce phase separation.The extract was centrifuged and the upper layer was filtered by microporous membrane.The cosmetic samples were separated on an Agilent Poroshell 120 EC-C18 column(4.6 mm × 100 mm,2.7 μm) by isocratic elution with acetonitrile-water(30:70,V : V) as mobile phases at a flow rate of 0.5 ml/min.The wavelength of detection was 210 nm and the external standard method was used for quantitation.The suspected samples were further verified by high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS).Results Good linearity was observed when the concentration of bimatoprost was within0.5 μg/ml-150 μg/ml,with correlation coefficients(r) 0.999 96.The limits of detection(LOD) was 1 mg/kg.The spiked recoveries were within 94.2%-103.5%,with the relative standard deviation(RSD) of 1.4%-3.0%.Eyelash enhancing cosmetic products from ten different manufactures were surveyed and bimatoprost was detected in three batches,with the content of 0.026%-0.030%.Conclusion This method can be applied successfully to the quality control in eyelash enhancing cosmetics.
