摘  要:目的研究松果菊苷抑制阿尔茨海默病转基因小鼠脑内氧化应激并减少β-淀粉样蛋白含量的作用。方法实验小鼠分为APP/PS1转基因小鼠(AD模型鼠)和松果菊苷治疗组。对照组给予生理盐水,治疗组小鼠给予松果菊苷(20 mg/kg/天)2个月。应用水迷宫实验方法检测两组小鼠的学习记忆能力;利用免疫荧光技术检测Aβ在AD模型鼠及松果菊苷治疗组小鼠脑内的变化情况;并考察氧化应激相关指标的变化。结果松果菊苷处理之后的AD模型小鼠脑内Aβ含量与AD组相比明显减少。氧化应激结果显示,松果菊苷干预处理后,小鼠脑内ROS含量降低,SOD活性明显升高。行为学结果提示松果菊苷治疗组小鼠学习记忆能力明显改善。结论松果菊苷可通过抑制APP/PS1转基因小鼠脑内氧化应激进而减少AD小鼠脑内的Aβ沉积数量,最终改善AD模型鼠的学习记忆能力,提示松果菊苷在AD的临床治疗方面具有重要的理论意义。Objective To mainly study the effect on the oxidative stress(OS)and the expression of amyloid β-protein(Aβ)in the brain of transgenic mice.Methods The mice were divided into two groups,namely the APP/PS1 Tg mice(AD model mice)and the echinacea treatment group.The mice were given echinacea treatment for 2 months,and the AD model mice(control)were given the saline.Morris water maze was used to investigate the learning and memory function.The expression of Aβ in the brain by immunofluorescence in AD model mice and echinacea treatment group,and the changes of OS related indexes were also investigated.Results We found that the activity and content of Aβ in the echinacea administration mice brain was decreased compared to the AD model mice.The OS results showed that echinacea could obviously inhibit the ROS and GSH,and promote the expression of SOD.Additionally,echinacea could improve the mice learning memory ability obviously.Conclusion Echinacea could decreased Aβ in AD mice brain through suppressing the level of OS in APP/PS1 transgenic mice brain,and improve the learning and memory ability,these results suggested that echinacea has important theoretical significance in the clinical treatment of AD.
