摘  要:目的探讨巴特日-7对输卵管炎大鼠抗炎作用的机制。方法将大鼠分为空白对照组、模型组、阳性对照组、实验组,每组10只。混合菌株接种法建立输卵管炎模型。阳性对照组灌胃金刚藤胶囊0.4 g·kg^(-1),实验组灌胃巴特日-7(0.5 g·kg^(-1)),空白对照组和模型组灌胃蒸馏水,每日1次,连续给药4周。用酶联免疫吸附(ELISA)法检测输卵管-子宫角组织肿瘤坏死因子-α(TNF-α)、白细胞介素-2(IL-2)、白细胞介素-6(IL-6)水平,用实时荧光定量逆转录聚合酶链反应(qRT-PCR)法检测输卵管-子宫角组织TNF-α、转化生长因子-β(TGF-β)基因表达。结果空白对照组、模型组、阳性对照组、实验组TNF-α基因表达值分别为0.53±0.08,0.99±0.14,0.67±0.06,0.62±0.07,TGF-β基因表达值分别为0.41±0.06,0.93±0.10,0.59±0.09,0.45±0.08,模型组较空白对照组上调,阳性对照组和实验组较模型组下调,差异均有统计学意义(均P<0.01)。结论巴特日-7通过下调TNF-α、TGF-β基因表达,阻断了TGF-β信号转导通路,从而延缓并抑制输卵管炎的形成。Objective To investigate the anti-inflammatory mechanism of Batari-7 on salpingitis rats.Methods The rats were divided into blank control group,model group,positive control group and experimental group,with 10 rats in each group.The salpingitis model was established by inoculation of mixed strains.The positive control group was given Kumgang Capsule(0.40 g·kg^(-1))by gavage,experimental group was given Batari-7(0.50 g·kg^(-1))by gavage,blank control group and model group was given distilled water,once a day,for 4 weeks.Enzyme linked immunosorbent assay(ELISA)method was used to detect the levels of tumor necrosis factor-α(TNF-α),interleukin-2(IL-2)and interleukin-6(IL-6).The real-time quantitative polymerase chain reaction(qRT-PCR)method was used to detect the expression of TNF-αand transforming growth factor-β(TGF-β)genes in the tubal and uterine tissues.Results The expression values of TNF-αgene in blank control group,model group,positive control group and experimental group were 0.53±0.08,0.99±0.14,0.67±0.06 and 0.62±0.07,TGF-βgene expression values were 0.41±0.06,0.93±0.10,0.59±0.09 and 0.45±0.08,model group was higher than blank control group,positive control group and experimental group were down-regulated compared with model group,all with significant difference(all P<0.01).Conclusion Batari-7 delayed and inhibited the formation of salpingitis by down-regulating the expression of TNF-αand TGF-βgenes and blocking the TGF-βsignal transduction pathway.
