摘  要:目的:基于"成分-靶点-通路"网络多层次挖掘夏枯草治疗视神经病变的潜在作用机制。方法:TCMSP中,以口服生物利用度大于30%和类药性大于0.18筛选出夏枯草中的药效成分,然后利用PubChem数据库筛选药效成分的潜在靶点,GenCards数据库筛选缺血性视神经炎、外伤性视神经病和Leber遗传性视神经病变相关的基因。采用Cytoscape3.7.0软件构建药效成分-靶点、疾病靶点PPI网络和疾病-成分-靶点网络。通过网络分析发现夏枯草治疗视神经病变的潜在关键成分和潜在作用靶点。随后对潜在作用靶点进行GO功能富集分析和KEGG通路富集分析,并构建成分-靶点-通路网络。结果:研究得到9个潜在药效成分,包括槲皮素、木犀草素、花翠素、桑色素、山奈酚、β-谷甾醇、菠菜甾醇、豆甾醇、豆甾醇葡萄糖苷,和13个潜在作用靶点包括ABCA1、AKT1、APOB、APOE、CASP3、CAT、IL6、PIK3CA、TNF、TP53、CCND1、PSAP、SNCA;GO功能和KEGG通路富集分析结果显示这13个作用靶点主要涉及35个生物过程通路,与6条代谢通路相关。结论:基于网络药理学的研究表明,夏枯草通过多成分-多靶点干扰多条信号通路发挥治疗视神经病变的效果,为阐述夏枯草治疗视神经病变潜在作用机制提供理论依据。Objective:To investigate the potential action mechanism of Prunella vulgaris in the treatment of optic neuropathy based on"components-targets-pathways"network.Methods:TCMSP database was used to analyze the effective components with OB(oral bioavailability)>30%and DL(drug-like)>0.18 as principles,PubChem database was used to screen potential targets for effective components,GenCards database was used to screen genes associated with ischemic optic neuritis,traumatic optic neuropathy and Leber hereditary optic neuropathy.Cytoscape3.7.0 software was used to construct the effective components-targets,disease targets PPI network and diseases-components-targets network.Through network analysis,the potential key components and potential targets of Prunella vulgaris in the treatment of optic neuropathy were found.GO functional analysis and KEGG pathway enrichment analysis were carried out for potential targets,and components-targets-pathways network was constructed.Results:Nine potential effective compounds of Prunella vulgaris were obtained,including quercetin,luteolin,delphinidin,morin,kaempferol,β-osterol,spinaterol,stigmasterol,stigmasterolglucoside.Thirteen potential action targets were obtained,including ABCA1,AKT1,APOB,APOE,CASP3,CAT,IL6,PIK3 CA,TNF,TP53,CCND1,PSAP,SNCA.The enrichment analysis results of GO function and KEGG pathway showed that these 13 action targets mainly involved 35 biological process pathways and were related to 6 metabolic pathways.Conclusion:The study based on network pharmacology shows that Prunella vulgaris can interfere with multiple signal pathways through multicomponents-multitargets to play a role in the treatment of optic neuropathy,which provides theoretical basis for the explanation of the potential mechanism of Prunella vulgaris in the treatment of optic neuropathy.
