摘  要:目的:通过网络药理学探究麻黄-苍术治疗新型冠状病毒肺炎(COVID-19)的作用靶点及信号通路,阐述其作用机制。方法:利用TCMSP、BATMAN-TCM数据库和相关文献筛选出麻黄-苍术的活性成分及靶标;通过GeneCards数据库筛选2019-nCoV的预测靶点;将两者靶点进行映射;利用Cytoscape3.7.2数据库构建活性成分-靶点网络图并筛选出核心靶点;再通过GOEAST、DAVID分别进行GO生物过程、KEGG通路富集分析。结果:根据筛选条件共获得麻黄-苍术活性成分32个,潜在作用靶点216个;与2019-nCoV共同靶点48个,关键靶点10个;获得1303条GO生物过程,在细胞生物过程正调控、生物节律调节、细胞对刺激的反应等方面靶点富集较集中;56条KEGG信号通路,靶点在甲型流感、百日咳、肺结核、乙型肝炎等通路上富集较多。结论:麻黄-苍术通过多靶点、多通路起到抗病毒、抑菌退热、止咳化痰、调节免疫的作用,最终达到治疗COVID-19的效果。Objective:To explore the targets and signaling pathways of Ephedrae Herba-Atractylodis Rhizoma in the treatment of novel coronavirus pneumonia(COVID-19)by network pharmacology,to expound its mechanism.Methods:The TCMSP,BATMAN-TCM databases and related literatures were used to screen out the active components and targets of Ephedrae Herba-Atractylodis Rhizoma;The GeneCards database was used to screen the predicted targets of 2019-nCoV;Two targets were mapped;The Cytoscape3.7.2 database was used to construct the network diagram of active components-targets and screen out the core targets;The GO biological process,KEGG pathway enrichment analysis were carried out by GOEAST and DAVID,respectively.Results:According to the screening conditions,32 active components of Ephedrae Herba-Atractylodis Rhizoma and 216 potential targets were obtained.48 common targets related with 2019-nCoV were obtained,10 key targets and 1303 GO biological processes were obtained.The targets were concentrated in the positive regulation of cellular biological processes,the regulation of biological rhythms and the response of cells to stimuli.56 KEGG signaling pathways were obtained and the targets were rich in influenza a,pertussis,tuberculosis,hepatitis b and other pathways.Conclusion:Ephedrae Herba-Atractylodis Rhizoma plays the roles of antivirus,bacteriostatic and antifebrile,cough and phlegm relieving,immune regulation through multiple targets and multiple pathways and finally achieves the effect of treating COVID-19.
