摘  要:目的探讨常用吸入麻醉药物七氟醚对老年大鼠相关认知功能影响及其对海马区Erk1/2/CREB/BDNF信号通路表达影响。方法30只20月龄的老年雄性wister大鼠,随机分为3组:对照组(C组n=10),七氟醚1组(S1组n=10),和七氟醚2组(S2组n=10),老年雄性wister大鼠放置一密闭麻醉箱中,该麻醉箱放置于预设温度为35℃度的水浴箱中。麻醉箱进气口与装有七氟醚气体挥发罐的麻醉机连接,出气口接延长管置于通风厨,S1组大鼠用1.5%七氟醚麻醉2h,S2组大鼠用3.0%七氟醚麻醉2h,C组大鼠暴露于纯氧中2h。麻醉气体由纯氧通过麻醉机输送通过该麻醉箱,实验中使用麻醉气体监测仪监测麻醉箱出气口的麻醉气体浓度。在麻醉过程中每隔30 min监测大鼠肤色,并记录呼吸频率。对照组在手术后第一天处死,七氟醚组分别于处理后第2天各断头处死10只,冰盘上快速取海马,Western Blot法测定海马区BDNF、CREB及Erk1/2相关表达,并分别在处死前2小时进行水迷宫实验,并判断其记忆认知相关行为是否产生了改变。结果试验组吸入低浓度1.5%七氟醚老年雄性wister大鼠试验后认知记忆功能不如对照组,试验组Western Blot法测定海马BDNF、CREB及Erk1/2相关表达均低于对照组,实验组组间比较浓度越高该信号通路表达活性越低。七氟醚预处理后吸入低浓度1.5%七氟醚后老年雄性wister大鼠认知功能减退,其机制可能与Erk1/2/CREB/BDNF信号通路表达有关,高浓度组(吸入低浓度3%七氟醚)影响情况较低浓度组显著。S1组、S2组麻醉后潜伏期均较麻醉前及C组麻醉后明显延长(P<0.05),S2组麻醉后潜伏期较S1组延长更为显著(P<0.05);S1及S2组大鼠120S内穿越原平台区域进入次数麻醉后显著低于麻醉前及C组,组间比较S2组<S1组,差异有统计学意义(P<0.05)。结论七氟醚可通过调节Erk1/2/CREB/BDNF信号通路的分子机制上对大鼠行为记忆认知功能产生影响从而加重老年Objective To investigate the effect of sevoflurane on cognitive function and the expression of ERK1/2/CREB/BDNF signaling pathway in hippocampus of elderly rats.Methods Thirty 20-month-old male Wister rats were randomly divided into three groups:control group(group C,n=10),sevoflurane group 1(group S1,n=10),and sevoflurane group 2(group S2,n=10).The old male Wister rats were placed in a closed anesthesia chamber in a water bath with a preset temperature of 35℃.The air inlet of the anesthesia box was connected with the anesthesia machine equipped with sevoflurane gas evaporation tank,and the air outlet was connected with an extension tube and placed in the ventilation kitchen.Rats in the S1 group were anesthetised with 1.5%sevoflurane for 2h,rats in the S2 group were anesthetised with 3.0%sevoflurane for 2h,and rats in the C group were exposed to pure oxygen for 2h.The anesthetic gas is conveyed by pure oxygen through the anesthesia machine and the anesthetic gas monitor is used to monitor the concentration of the anesthetic gas at the air outlet of the anesthesia box.The skin color of the rats was monitored every 30 min during anesthesia,and the respiratory rate was recorded.The control group was killed on the first day after surgery,and 10 animals in the sevoflurane-treated group were killed on the second day after treatment,respectively.The hippocampi were quickly collected from the ice plate,and the expressions of BDNF,CREB and ERK1/2 in the hippocampi were determined by Western Blot.The water maze experiment was conducted 2 hours before the death to determine whether the memory and cognition related behaviors were changed.Results The cognitive and memory function of the experimental group after inhalation of low concentration 1.5%sevoflurane was worse than that of the control group.The related expressions of BDNF,CREB and ERK1/2 in the hippocampus of the experimental group were lower than those of the control group by Western Blot.The higher the concentration of the experimental group,the lower the expressi
