摘 要:为了对疑似禽博尔纳病毒感染病例进行确诊,并了解鹦鹉源禽博尔纳病毒毒株的流行特点,本试验设计合成1对特异性引物,对疑似感染禽博尔纳病毒而死亡的鹦鹉进行检测,并设计7对首尾重叠的引物对其中1株确诊的博尔纳病毒全基因进行RT-PCR扩增、测序,应用DNAStar和MEGA6.06软件将本次获得的PaBV-4与GenBank上公布的PaBV序列进行同源性和遗传进化分析。结果显示,死亡的10只鹦鹉中,有5只感染PaBV-4,1只感染PaBV-5。选取其中1份PaBV-4组织样品做全基因测序和序列分析,结果显示PaBV-4基因组全长8915 nt,编码6种蛋白,与NM20分离株核苷酸相似性最高(99.6%),与6758分离株氨基酸相似性最高(99.8%);遗传进化分析显示,与PaBV-4各分离株M14、AG5、6758亲缘关系最近,同属一个小的分支,其次是NM01,与PaBV-5各分离株亲缘关系最远。In order to confirm the suspected cases of avian Bornavirus infection and understand the epidemic characteristics of the avian Bornavirus strains from parrots,a pair of specific primers were designed and synthesized to detect dead parrots suspected of being infected with avian Bornavirus.Seven pairs of overlapping primers were designed to perform RT-PCR amplification and the whole genome of one of the confirmed Bornavirus strains was sequenced,and DNAStar and MEGA6.06 software were used to analyze the homology and genetic evolution between the PaBV-4 obtained this time and the PaBV sequence published on GenBank.The results showed that among the ten dead parrots,five were infected with PaBV-4 and one was infected with PaBV-5.One of the PaBV-4 tissue samples was selected for full genome sequencing and sequence analysis.The results showed that the full-length genome of PaBV-4 was 8915 nucleotides(nts),encoding 6 proteins,and it has the highest nucleotide similarity to the NM20 isolate,which is 99.6%.Its amino acid homology with isolate 6758 was the highest(99.8%).Genetic evolution analysis showed that it was closely related to PaBV-4 isolates M14,AG5,6758,and belonged to a small branch,followed by NM01,and it had the farthest relationship with each isolate of PaBV-5.
