摘  要:以50,100,150μmol/L二烯丙基二硫化物(diallyl disulfide, DADS)处理MDCC-MSB-1细胞24 h后,利用吖啶橙(acridine orange, AO)染色法对细胞凋亡的形态进行初步观察;Annexin V-FITC/PI双染法检测细胞凋亡率;JC-1染色法检测线粒体膜电位的变化;Western blot检测P53的蛋白表达;蛋白活性检测试剂盒检测Caspase-3和Caspase-9的活性变化;Q-PCR检测Caspase-3和Caspase-9 mRNA的表达情况。结果显示,与空白组相比,DADS诱导MDCC-MSB-1细胞发生凋亡形态变化,增加细胞凋亡率,同时诱导p53蛋白表达量和线粒体膜电位下降,上调Caspase-3和Caspase-9基因表达水平和蛋白活性。MDCC-MSB-1 cells were treated with 50,100,150 μmol/L diallyl disulfide(DADS) for 24 h, and the apoptotic morphology of cells was observed by staining with acridine orange(AO);cell apoptosis rates were detected by Annexin V-FITC/PI double staining;changes in mitochondrial membrane potential were detected by JC-1 staining;the expression of p53 protein was detected by Western blot;the protein activity of Caspase-9 and Caspase-3 was measured by protein activity assay kit;the mRNA expression levels of Caspase-9 and Caspase-3 were analyzed by q-PCR.The results showed that compared with the blank group, after treatment with DADS,the cytoplasm of MDCC-MSB-1 cells changed from homogenization to fragmentation, and cell apoptotic rate up-regulated at the same time, the p53 expression and mitochondrial membrane potential decreased, the gene expression and protein activity of Caspase-3 and Caspase-9 increased.This study laid a theoretical foundation for further study on the use of DADS to combat Marek’s disease.
